Conclusion Humoral and cellular immune responses developed in mice after intranasal delivery of model mouse polyomavirus derived VLPs carrying epitope of enhanced green fluorescence protein (EGFP) Model chimeric EGFP-VLPs were purified and used for immunization of mice. Immune response of immunized animals was examined. No specific antibodies against EGFP protein, but high titers of specific antibodies against major structural protein VP1 were developed in the sera of immunized animals. Splenocytes derived from immunized animals secreted IL-2 and IFN-γ after their antigen (EGFP or VP1) restimulation. Proliferation of CD4+, but not CD8+ T cells from immunized mice after the stimulation with both EGFP and VP1 was observed. No EGFP specific cytotoxic activity of splenocytes from immunized mice was detected. The presentation of EGFP-VLPs in the context ofMHC class II was blocked by inhibitors of endo-lysosomal as well as proteasomal compartments. Changes in the numbers of CD25+Foxp3+ subpopulation of CD4+ T cells were observed in the spleens if immunized mice. Shrnutí Humorální a buněčná odpověď myší na intranasální aplikaci modelových polyomavirových VLPs nesoucích zelený fluorescenční protein Imunizovali jsme myši modelovými chimerickými EGFP-VLPs a studovali jsme jejich imunitní odpovědi. Detekovali jsme protilátky proti proteinu VP1, ale ne protilátky proti proteinu EGFP v sérech imunizovaných myší. Prokázali jsme produkci IL-2 a IFN u splenocytů z imunizovaných myší. Pozorovali jsme proliferaci CD4+, ale ne CD8+ T buněk, specifickou na VP1 i na EGFP. Nepodařilo se nám prokázat cytotoxickou aktivitu specifickou proti EGFP. Presentace EGFP-VLPs v souvislosti s MHC II třídy byla blokována inhibitory endolysosomálních i proteasomálních kompartmentu. Zaznamenali jsme změny v počtech CD25+Foxp3+ subpopulací CD4+ T buněk ve slezinách imunizovaných myší. Chimerické VLPs odvozené od myšího polyomaviru nesoucí epitopy lidského fúzního proteinu Bcr-Abl (Bcr-Abl VLPs) Na základě modelových EGFP-VLPs jsme připravili chimerické Bcr-Abl VLPs nesoucí 171 aminokyselinovou sekvenci z oblasti zlomu lidského fúzního proteinu Bcr-Abl. Bcr-Abl VLPs jsme použili k imunizaci myší a studovali jsme jejich imunitní odpovědi. Prokázali jsme vysoký titr protilátek proti VP1 v sérech imunizovaných myší, avšak protilátky proti Bcr-Abl jsme nebyli schopni detekovat. Bcr-Abl VLPs aktivovaly...
