Od objevu volné DNA (cfDNA) uplynulo více než 70 let, ale k největšímu zájmu o toto téma a k poznání dochází bezesporu v posledních třiceti letech. Využití nachází především v onkologii a prenatální diagnostice. Zatímco v těchto oborech je rutinně diagnosticky využívána, jen málo se ví o jejích fyziologických funkcích v organismu. Náš výzkum se proto zaměřuje na pochopení této úlohy a v experimentech pracuje zejména se vzorky získanými od zdravých jedinců. První studie byla zaměřena na rozdíly mezi plazmou a sérem zdravých jedinců. Položili jsme si otázku, zda se u nich liší koncentrace cfDNA a zastoupení telomerických sekvencí a zjistili jsme, že sérum obsahuje signifikantně více cfDNA než plazma, na druhou stranu je plazma relativně bohatší na telomerické sekvence. Při stimulačních experimentech s THP1 buňkami vykazovaly vzorky kultivované se sérem ošetřeným DNázou (bez přítomnosti cfDNA) vyšší expresi mRNA TNF-α (Tumor necrosis factor α), než vzorky kultivované s neošetřeným sérem a stejný trend byl pozorován i při stimulaci se vzorky plazmy. Studie zahrnující vzorky plazmy od deseti pacientů s celiakií a deseti zdravých kontrol ukázala při stimulaci THP1 buněk signifikantní rozdíly exprese mRNA TNF-α u DNázou ošetřených vs. neošetřených vzorků, kdy u ošetřených vzorků plazmy byla jejich... More than 70 years have passed since the discovery of cell-free DNA (cfDNA), but the greatest interest in this topic and knowledge has undoubtedly occurred in the last thirty years. It is used mainly in oncology and prenatal diagnostics. While it is routinely used diagnostically in these fields, little is known about its physiological functions in the organism. Our research therefore focuses on understanding this role and in experiments works mainly with samples obtained from healthy individuals. The first study focused on the differences between plasma and serum in healthy individuals. We asked the question whether they differ in cfDNA concentration and telomeric sequences abundance. We found that the serum contains significantly more cfDNA than plasma, on the other hand, plasma is relatively richer in telomeric sequences. In stimulation experiments with THP1 cells, samples cultured with DNase-treated serum (without cfDNA) showed a higher expression of mRNA TNF-α (Tumor necrosis factor α) than samples untreated. The same trend was observed when plasma samples were stimulated.
