Galina Karabanovich, Ph.D. Student: Pavel Kruchkou Název diplomové práce: Syntéza analog chelátoru SIH se zvýšenou stabilitou vůči hydrolýze Nadměrné množství volných iontů železa v organismu může katalyzovat tvorbu hydroxylových radikálů, které se podílí na rozvoji různých onemocnění. Jednou z možností ochrany organizmu je chelatace volných iontů železa pomocí chelátorů, a tím zabránit rozvoji železem katalyzovanému oxidačnímu stresu. Účinnými chelátory jsou látky ze skupiny analogů salicylaldehyd isonikotinoyl hydrazonu. Salicylaldehyd isonikotinoyl hydrazon (SIH) je chelátor, který vytváří komplexy s ionty Fe 3+ . Díky jeho vysoké lipofilitě může být podáván perorálně. Jeho hlavní nevýhodou je nízká stabilita ve vodném prostředí způsobená rychlou hydrolýzou hydrazonové vazby. Cílem této práce bylo syntetizovat série nových aroylhydrazonů odvozených od chelátoru SIH, které by měly mít zlepšenou schopnost chelatace iontů železa a s ní spojenou schopnost ochrany buněk před oxidačním stresem, a potenciálně také zvýšenou stabilitu vůči hydrolýze. One of the ways how to protect the body is to chelate free iron ions with chelators, in order to prevent the development of iron- catalyzed oxidative stress. Effective chelators are substances from the group of salicylaldehyde analogues of isonicotinoyl hydrazone. Salicylaldehyde isonicotinoyl hydrazone (SIH) is a chelator that forms complexes with Fe 3+ ions. Due to its high lipophilicity can be administered orally. Its main disadvantage is the low stability in the aqueous environment caused by the rapid hydrolysis of the hydrazone bond.
