Kontaktní zóny cytotypů nám poskytují jedinečnou příležitost pro studium dynamiky a evoluce polyploidních komplexů. Butomus umbellatus (šmel okoličnatý) je jedním z mála druhů, u kterých se přirozeně vyskytuje diploidní a triploidní cytotyp. Během předchozích průzkumů na jihovýchodním Slovensku byly odhaleny první doposud známé cytotypově smíšené populace B. umbellatus. Hojný výskyt druhu v oblasti s do značné míry přirozenou dynamikou vodních ekosystémů je unikátním modelovým systémem, který umožňuje nezkreslené srovnání vlastností a genetické diverzity rostlin obou cytotypů v přirozených podmínkách včetně možnosti jejich šíření v krajině. Cílem práce je objasnit evoluční procesy probíhající v kontaktní zóně diploidního a triploidního cytotypu B. umbellatus na jihovýchodním Slovensku. Dále pomocí průtokové cytometrie a molekulárně genetických metod studia zjistit, zda se jedná o primární nebo sekundární kontaktní zónu cytotypů, a určit míru genetické variability obou cytotypů na vnitropopulační a mezipopulační úrovni v zájmové oblasti i v širším geografickém kontextu. Pomocí analýzy mikrosatelitových lokusů bylo zjištěno, že kontaktní zóna cytotypů je jak primárního, tak sekundárního charakteru a mezi diploidy a triploidy B. umbellatus dochází ke genovému toku i přes ploidní bariéru. Tyto závěry byly... Contact zones of cytotypes harbour a unique opportunity to study the dynamics and evolution of mixed-ploidy systems. Butomus umbellatus (flowering rush) is one of a few species in which diploid and triploid cytotype is present in nature. The first known mixed-ploidy populations of B. umbellatus were discovered in south-eastern Slovakia during the previous studies. Common occurrence of the species in an area with substantially natural dynamics of wetland ecosystems represents a unique model system for unbiased comparisons of traits and genetic diversity between diploid and triploid individuals along with their dispersal capabilities on a landscape level. The main aim of of this thesis is to uncover the truth behind evolutionary processes that are taking place in the contact zone of diploid and triplioid cytotype of B. umbellatus in south-eastern Slovakia. Using flow cytometry and molecular methods (microsatellites) we will test whether this is a primary or secondary contact zone and assess the level of genetic variability between individuals and populations of both cytotypes.
