Úvod: Nové syntetické drogy (NSD) se s přelomem tisíciletí staly populární složkou drogové scény nejen v České republice, ale i po celém světě. Hlavním důvodem jejich oblíbenosti a rozšíření je jejich legálnost a záliba uživatelů vyhledávat nové zážitky. Dochází tak k neustále novému produkování substancí s nejrůznějšími chemickými modifikacemi struktury, avšak odborné informace o akutních a chronických účincích, farmakologii a toxikologii převážně chybí nebo jsou nedostačující. Cíl: Hlavním cílem disertační práce bylo studium a evaluace NSD z řad katinonů. Dílčí cíle byly následující: 1) zhodnotit míru intenzity stimulačních účinků 2) zhodnotit míru narušení senzomotorického zpracování 3) zhodnotit akutní toxicitu z hlediska rizika hypertermie 4) zhodnotit návykový potenciál. Metodika: Testované NSD v rámci této práce byly mefedron (2.5, 5, 20 mg/kg) metylon 5, 10, 20, 40 mg/kg) a nafyron (5, 10, 20 mg/kg). Všechny NSD byly aplikovány subkutánně v objemu 2 ml/kg. Dospělí samci laboratorního potkana kmene Wistar byli behaviorálně testováni v testu otevřeného pole (n = 10/skupina), testu prepulzní inhibice (n = 10/skupina) a testu podmíněné preference místa (n = 13/skupina). Pro farmakokinetické experimenty byla potkanům odebrána krev a orgány (mozek, plíce, játra) v předem stanovených intervalech,... Introduction: Since the beginning of this century, new synthetic drugs (NSD) have become a popular component of Czech Republic and worldwide drug scene. The main reason for their popularity and prevalence is their legality and the interest of users to seek new experiences. This leads to a constant production of substances with various chemical structure's modifications, but information about acute and chronic effects, pharmacology and toxicology are mostly missing or they are not sufficient. Aims: The main aim of this doctoral thesis was to evaluate NSD from the ranks of cathinones. The subobjectives were as follows: 1) evaluation of stimulation potential 2) evaluation of potency to disrupt sensorimotor gating 3) evaluation of acute toxicity in terms of the risk of hyperthermia 4) evaluation of addictive potential. Methods: The NSDs tested in this thesis were mephedrone (2.5, 5, 20 mg/kg), methylone 5, 10, 20, 40 mg/kg) and naphyrone (5, 10, 20 mg/kg). All NSDs were administered subcutaneously in a volume of 2 ml/kg.
