Nádorová onemocnění jsou po celém světě jednou z nejčastějších příčin úmrtí. V České republice je právě pětina úmrtí zapříčiněna onkologickým onemocněním. Velkým problémem jsou při léčbě onkologických onemocnění nežádoucí účinky chemoterapie, které činí pacientům léčbu ještě těžší. V případě předkládané bakalářské práce je výzkum soustředěn na syntézu polymerních nosičů, které by mohly sloužit jako dopravci cytostatik do nádorové tkáně. Při použití nosičů cytostatik by bylo působení na zdravé tkáně a tím způsobené nežádoucí účinky výrazně sníženy. Práce je zaměřená na syntézu a fyzikálně chemickou charakterizaci nových diblokových polymerních nosičů cytostatik, konkrétně cytostatika docetaxelu. V předkládané práci je vedle úspěšné syntézy nových vodorozpustných polymerních prekurzorů a konjugátů na bázi N-(2-hydroxypropyl)methakrylamidu studována hydrolytická stabilita syntetizovaných diblokových systémů v 0,3M fosfátovém pufru o pH 7,4 simulujícím prostředí krevního řečiště. Úspěšně bylo ověřeno, že syntetizované polymerní systémy jsou hydrolyticky labilní, a tudíž snadno vylučitelné z organismu. V práci je dále prokázaný vliv typu oxokyseliny použitých pro modifikaci cytostatika na rychlost uvolňování neseného cytostatika z polymerního nosiče. Na závěr práce byla studována in vitro cytotoxicita... Cancer diseases are the most common cause of death over the world. One fifth of death is caused by cancer in the Czech Republic. The big problem in the treatment of cancer diseases are side effects of chemotherapy, which make treatment more difficult for patients. The present bachelor thesis is focused on synthesis and characterization of novel polymer carriers. Polymer carriers can target the cytostatic drug to the tumor tissue and minimize side-effect of chemotherapies. This bachelor thesis is focused on synthesis and physical chemistry characterization of new diblock polymer carriers, specifically carries of docetaxel. Successful synthesis of novel water-soluble polymer precursors and conjugates based on N-(2-hydroxypropyl)- methacrylamide is described and discussed in this thesis. The hydrolytic stability of the synthesized diblock systems was studied in 0,3M phosphate buffer at pH 7,4 simulating the bloodstream environment. It has been successfully verified that the synthesized polymer systems are hydrolytically degradable after fulfilling its role of drug carrier and therefore can be finally excreted from the organism.
