RNDr. Veronika Opletalová, Ph.D. Název dizertační práce: In silico studium interakcí cholinesteras s jejich modulátory a návrh nových látek tohoto typu Počítačové modelování je významným nástrojem současného vědeckého výzkumu. Umožňuje studovat strukturu, konformace, dynamiku a vzájemné nevazebné interakce biologického systému. Hlavním cílem je nalezení energeticky nejvýhodnější konformace pomocí minimalizace energie systému. Z takto vypočtených hodnot můžeme předpovědět ztrátu aktivity pro určité analogické sloučeniny, což umožňuje snížit počet látek, které by bylo zapotřebí následně syntetizovat. Molekulové modelování se podílí na popisu interakcí ligandů s většími systémy a zlepšuje tak orientaci v milionech aktivních látek, které by mohly být využity jako potenciální léčivo. Stává se tak významnou součástí farmaceutického průmyslu. Cílem této práce bylo ověřit vybranou metodiku počítačové predikce ligand-protein komplexů, pomocí molekulárního dockingu získat popis základních interakcí modulátorů cholinesteras s těmito enzymy a na základě těchto poznatků navrhnout strukturu nového účinného inhibitoru acetylcholinesterasy. It allows to study the structure, conformation, dynamics and mutual interaction of nonbinding interactions of a biological system. The main objective is to find the best energy conformations by minimizing the energy of the system. On the base of these calculated values, one can predict the loss of activity for certain analogous compounds, thus reducing the number of materials that would be needed to subsequently synthesize. A molecular modeling contributes to describe the interactions of ligands with larger systems and improves the orientation in millions of active substances that could be used as potential drug. It becomes an important part of the pharmaceutical industry.
