PharmDr. Lucie Nováková, Ph.D. Název diplomové práce: Vývoj UHPLC-MS/MS metody pro analýzu vybraných antivirotik v HILIC a RP módu Cílem této diplomové práce je vývoj a optimalizace metod pro analýzu vybraných antivirotik a zároveň srovnání vhodnosti módu RP a HILIC pro tuto aplikaci. Vybraná antivirotika jsou užívána v terapii HIV a hepatitidy B a/nebo C. Analýzy proběhly s využitím systému ACQUITY UPLC I-Class PLUS s hmotnostním detektorem typu trojitého kvadrupólu. Nejprve byla optimalizována MS/MS metoda. Pro RP mód bylo testováno celkem 6 stacionárních fází (BEH Shield RP18, BEH Phenyl, BEH C18, CSH Fluoro-phenyl, CSH Phenyl-hexyl a CSH C18) a 4 fáze mobilní (acetonitril v gradientové eluci s vodnou složkou: kyselina mravenčí, kyselina octová, mravenčan amonný pH 3 a octan amonný pH 9). Podmínky separace byly nastaveny následovně: teplota 40 řC, průtok 0,3 mL/min, gradientová eluce od 5 % ACN do 98 % ACN do 5. minuty, s izokratickým krokem na konci analýzy. Pro mód HILIC bylo testováno 10 stacionárních fází (BEH Amide, BEH HILIC, Cortecs HILIC, Ascentis Express HILIC, Ascentis Express OH5, Syncronis HILIC, Kinetex HILIC, Luna HILIC, Luna Omega SUGAR a Zorbax HILIC plus) a... Targeted antivirals are used in treatment of HIV and hepatitis B and/or C. LC analyses were carried out on the ACQUITY UPLC I-Class PLUS system with triple quadrupole MS detector. Initially MS/MS method was optimized. In RP mode total of 6 stationary phases (BEH Shield RP18, BEH Phenyl, BEH C18, CSH Fluoro-phenyl, CSH Phenyl-hexyl, and CSH C18) and 4 mobile phases (ACN in gradient elution with aqueous component: formic acid, acetic acid, ammonium formate pH 3, ammonium acetate pH 9) were tested. Separation conditions were as follow: temperature 40řC, flow rate 0.3 mL/min, gradient elution from 5 % ACN to 98 % ACN within 5 minutes with isocratic step at the end of analysis.
