Juraj Martiška Předložená diplomová práce je zaměřena na biodegradovatelné polymerní nanočástice s terbinafinem na bázi kopolymeru kyseliny mléčné a kyseliny glykolové, větveného na kyselině polyakrylové. Pro přípravu nanočástic byla použita nanoprecipitační metoda a studován vliv formulačních faktorů na charakteristiky nanočástic. Sledovanými formulačními faktory byly koncentrace polymeru, množství terbinafinu, podíl vnitřní fáze a koncentrace tenzidu. Byly získány nanočástice o velikosti 120 nm až 300 nm v závislosti na podmínkách přípravy. Polydisperzita nanočástic byla ve všech případech pohybovala od 0,080 do 0,230. Připravené nanočástice byly stabilní, což dokazují hodnoty zeta potenciálu vyšší než 38 mV. Kladná hodnota zeta potenciálu je žádoucí z hlediska adheze na pokožku a sliznice při topické a oční aplikaci nanočástic s terbinafinem. Velikost částic byla ovlivněna zejména množstvím polyesteru použitého při přípravě. S rostoucí koncentrací polyesteru ve vnitřní fázi rostla velikost nanočástic. Vliv terbinafinu a koncentrace... Juraj Martiška The diploma thesis is focused on biodegradable polymer nanoparticles loaded by terbinafine based on the copolymer of glycolic and lactic acid branched on polyacrylic acid. The nanoprecipitation method was employed, and the influence of formulation factors on nanoparticle characteristics was studied. The following formulation factors were the concentration of the polymer, the amount of terbinafine, and the concentration of surfactant. Nanoparticles of 120 nm to 300 nm were obtained depending on the preparation conditions. The nanoparticle polydispersity was in all cases from 0.080 to 0.230. The prepared nanoparticles were stable, as evidenced by zeta potential values above 38 mV. A positive zeta potential is desirable for dermal and mucosal adhesion in the topical and ocular application of nanoparticles with terbinafine.
