[cze][eng] Mitochondriální pór přechodné propustnosti (mPTP) je Ca2+ dependentním kanálem nacházejícím se ve vnitřní mitochondriální membráně, který je inhibován nanomolárními koncentracemi imunosupresiva cyklosporinu A. Společně s dalšími interagujícími proteiny, které regulují jeho otevření, utváří mPTP proteinový komplex přechodné propustnosti. Přetrvávající otevření mPTP je doprovázeno bobtnáním mitochondrií a následným kolapsem organely, jež předchází vyplavení proapoptotických proteinů a programované buněčné smrti. Kanál utvářející proteinová jednotka mPTP zůstává i přes intenzivní dlouholetý výzkum neznámá. Napříč historií zkoumání mPTP byl za kanál utvářející jednotku přijímán proteinový komplex ANT-VDAC, samotný protein ANT, PiC i ATP syntáza. I přes skutečnost, že nám doposud není známá přesná strukturní podstata, je mPTP přisuzována role v celé řadě onemocnění a patofyziologických stavů s nimi spojenými. Patří mezi ně zejména role v ischemickém/reoxidačním poškození, nervových a svalových dystrofiích i v nádorovém bujení. Klíčová slova: mitochondrie, mitochondriální pór přechodné propustnosti, cyklosporin A, programovaná buněčná smrt, ATP syntáza, oxidačně fosforylační aparát.Mitochondrial permeability transition pore (mPTP) is Ca2+ dependent channel localised in the inner mitochondrial membrane. One of its defining characteristics is inhibition by nanomolar concentrations of immunosuppressant cyclosporine A (CsA). Together with additional interacting proteins, which regulate its opening, mPTP forms a permeability transition protein complex. Persistent opening of mPTP is accompanied by mitochondrial swelling and a subsequent collapse of organelle, which precedes release of proapoptotic proteins and programmed cell death. Channel forming unit of mPTP remains unknown, despite intense and long-lasting study. Numerous proteins were proposed to play a role of channel forming subunit of mPTP, including complex of ANT and VDAC, ANT alone, PiC or even ATP synthase. Despite the fact, that molecular structure remains elusive, mPTP seems to play a role in a range of pathophysiological processes or diseases associated with them. Among others this includes ischemia/reperfusion injury, neurological and muscle dystrophies, or tumorigenesis.
