[cze][eng] Transkripční faktory z rodiny proteinů TEAD jsou důležitou součástí mnohých eukaryotických regulačních procesů. Jsou součástí Hippo signální dráhy a podílí se na kontrole diferenciace buněk a apoptózy. Zájem o tyto proteiny též souvisí s jejich spojením s onkologickými onemocněními. DNA-vazebná doména TEAD transkripčních faktorů interaguje s M-CAT elementy dvouvláknové DNA, kterými jsou i sekvence dsDNA z exonu a enhanceru genu C-MYC, jejichž interakce s DNA-vazebnou doménou proteinů TEAD byla v této práci studována především iontovou mobilitou ve spojení s hmotnostní spektrometrií za použití nativní ionizace nanoelektrosprejem. Studiu proteinu TEAD1-DBD a jeho dsDNA komplexů předcházela produkce DNA-vazebné domény TEAD1 dle již optimalizovaného protokolu. TEAD1-DBD a jeho komplexy byly dále studovány pomocí kolizně indukované aktivace a zahřívání sprejovaného roztoku při použití nativní hmotnostní spektrometrie ve spojení s iontovou mobilitou. Při kolizní aktivaci bylo pozorováno kolizně indukované rozbalování proteinu i obou jeho komplexů s DNA. Při zahřívání sprejovaného roztoku bylo sledováno tavení dsDNA i komplexů a z naměřených hodnot byly určeny teploty tavení jednotlivých vzorků. Pomocí kalibrace iontové mobility s putující vlnou byly určeny kolizní průřezy jednotlivých molekul.Transcription factors from TEAD protein family play an important role in various eukaryotic regulatory processes. They are partly responsible for cell differentiation and apoptosis due to their involvement in the Hippo signalling pathway. Scientific interest in this protein family is mainly because they are associated with carcinogenesis. DNA-binding domain of TEAD transcription factors interacts with M-CAT element of double-stranded DNA, which is present also in enhancer and exon of C-MYC gene. Interaction between TEAD1 DNA-binding domain and C-MYC dsDNA was investigated in this bachelor's thesis mainly by ion mobility in connection with mass spectrometry using native nESI ionization. For the further analysis of TEAD1-DBD and its complexes with dsDNA, the protein was firstly recombinantly expressed by the optimized protocol in our laboratory. Investigation of TEAD1-DBD and its complexes was done using two IM-MS activations. Firstly, the collision-induced unfolding was performed and afterwards perturbation of the sample by preheating nESI emitter was done. The first activation led to the collision- induced unfolding of some charge states of TEAD1-DBD protein and both protein-DNA complexes.
