Abstrakt: [cze][eng] Tato diplomová práce se zabývá stanovením glukosinolátů v rostlinném materiálu pomocí kapilární elektroforézy a vysokoúčinné kapalinové chromatografie. V práci je popsána chemická struktura glukosinolátů, biosyntéza a degradace glukosinolátů a také jsou popsány jejich biologické účinky. Část této práce se zabývá i metodami, kterými byly glukosinoláty v rostlinných materiálech stanoveny. V experimentální části je pak popsána samotná separace intaktních glukosinolátů pomocí kapilární elektroforézy a vysokoúčinné kapalinové chromatografie. Pro stanovení glukosinolátů byly k dispozici dva rostlinné materiály, a to lyofilizované listy řepky a šťáva z brokolice. Při stanovení intaktních glukosinolátů metodou kapilární elektroforézy se využila micelární elektrokinetická chromatografie za použití kationtového povrchově aktivního činidla. Po nalezení optimálních podmínek pro separaci intaktních glukosinolátů bylo zjištěno, že v rostlinných vzorcích je nemožné tyto látky stanovit. Důvodem byly interference z matrice, které toto stanovení rušily. Zatímco za použití vysokoúčinné kapalinové chromatografie se za optimálních podmínek, podařilo některé z intaktních glukosinolátů identifikovat v rostlinném vzorku řepky. Dále byly získány kalibrační závislosti jednotlivých glukosinolátů a vypočtena výtěžnost a...This diploma thesis deals with the determination of glucosinolates in plant material by capillary electrophoresis and high performance liquid chromatography. The chemical structure, biosynthesis, degradation, and also biological effects of glucosinolates are described. One part of this work also deals with the methods, which glucosinolates in plant materials were determined by. The experimental part describes the separation of intact glucosinolates by capillary electrophoresis and high performance liquid chromatography. Two plant materials were available for the determination of glucosinolates, namely lyophilized rapeseed leaves and broccoli juice. Micellar electrokinetic chromatography using a cationic surfactant was used to determine intact glucosinolates by capillary electrophoresis. After finding the optimal conditions for the separation of intact glucosinolates, it was found that it is impossible to determine these substances in plant samples. The reason was interference from the matrix, which interfered with this determination. While using high performance liquid chromatography under optimal conditions, some of the intact glucosinolates were identified in a rapeseed plant sample.
