Dysregulace NTs může vést k ovlivnění nukleosidové homeostázy s negativním dopadem na vývin placenty a plodu a může vést ke změně placentární farmakokinetiky léčiv. Proto je porozumění exprese a funkce NTs nutná pro účinnou a bezpečnou farmakoterapii v období těhotenství. Cílem bylo studovat regulační dráhy genové exprese koncentračního nukleosidového transportéru 2 (CNT2) aktivované adenylátcyklázou (AC). K tomu byly použity dvě metody, qRT-PCR a in vitro akumulační zkoušky využívající modelový substrát [3 H]-adenosin. Buněčná linie BeWo, odvozená od lidského placentárního choriokarcinomu, byla vystavena působení aktivátoru AC, forskolinu (50 μM) a/nebo inhibitorů signálních drah AC/cAMP/PKA a AC/cAMP/MAPK (MEK1/2 a p38 MAPK), tedy inhibitoru PKA, KT 5720 (5 μM), inhibitoru MEK1/2, U0126 (10 μM) a inhibitoru p38 MAPK, SB202190 (10 μM). Dysregulation of NTs can disrupt nucleoside homeostasis with a negative consequences on placental and fetal development and can lead to a change in placental pharmacokinetics of nucleoside-derived drugs. Therefore, understanding the expression and function of NTs is necessary for effective and safe pharmacotherapy during pregnancy. The aim of this diploma thesis was to study the adenylate cyclase (AC) activated regulatory pathways of gene expression of concentrative nukleoside transporter 2 (CNT2). For this purpose, qRT-PCR and in vitro accumulation assays using the model substrate [3 H]-adenosine were employed.
