Adheze mezi plasmatickou membránou a buněčnou stěnou a existence kontinua těchto dvou kompartmentů je důležitá pro přenos signálů, například při napadení patogenem, během expanze buněk a růstu buněčné stěny, či reakce na podmínky okolního prostředí. Tuto adhezi zajišťuje kromě turgoru i fyzické propojení obou kompartmentů. Jeden z nejznámějších příkladů fyzického propojení najdeme v kořenovém systému, v oblasti Casparyho proužku, kde je potřebné pro udržení apoplastické bariéry kořene, a to i za nepříznivých podmínek a následné plazmolýzy. O fyzickém propojení a účastnících se makromolekulách se toho do dnešní doby ví málo, jsou ale vytipováni kandidáti, kteří by se této interakce mohli účastnit. Tato diplomová práce se zabývá arabinogalaktanovými (AGP) proteiny s fasciclin-like doménou (konkrétně FLA9 a FLA2). Tyto proteiny by mohly hrát roli v adhesi plasmatické membrány a buněčné stěny a zároveň být zapojeny v regulaci růstu a vývoje kořenového systému, vzhledem k jejich relativně silné expresi v kořenech, především v elongační zóně a v primární kůře, včetně endodermis (dle čipových dat). Dále se práce v menší míře věnuje i integrin-like proteinu At14A. Nejzajímavějším výsledkem provedených experimentů je zjištění, že mutantní rostlina se sníženou expresí FLA9 genu má zhoršený růst kořenového... Adhesion between the plasma membrane and the cell wall and the existence of the continuum of these two compartments is needed for signal transmission, e.g. under pathogen attack, during cell expansion and cell wall growth, or in response to environmental conditions. This adhesion is, in addition to the turgor, provided by physical connection of both compartments. One of the best-known examples of physical connection is found in the root system, in the Caspary strip region, where it is required to maintain apoplastic barriers of the root system, even under adverse conditions and consequent plasmolysis. There is little information about the physical interconnection and the participating macromolecules but there are candidates, which could participate in this interaction. The diploma thesis deals with arabinogalactan (AGP) proteins with fasciclin-like domain (namely FLA9 and FLA2). These proteins may play a role in the adhesion of plasma membrane and cell wall and may be involved in the growth regulation and development of the root system. Both genes are are relatively strongly expressed in the roots, especially in the elongation zone and in the cortex, including the endodermis (according to chip data).
