PharmDr. Přemysl Mladěnka, Ph.D. Název diplomové práce: Chelatace železnatých iontů deriváty xanthen-3-onu Železo je esenciální prvek, důležitý pro správnou funkci buněk. Dysbalance hladiny železa může vést k závažným onemocněním. Jelikož neexistuje exkreční mechanismus, je jeho homeostáza regulována na úrovni vstřebáváním ve střevě. K nadbytku železa, který vede k poškození tkání z důvodu katalýzy tvorby volných radikálů, dochází z důvodu genetických onemocnění jako je hemochromatóza nebo při častém podávání transfúzí. Racionální terapií při přetížení železem je podávání jeho chelátorů. Cílem této studie bylo zhodnotit schopnost derivátů 2,6,7-trihydroxyxanthen-3-onu (syntetizovaných na Univerzitě v Sarajevu - Dr. Durić) chelatovat železo ve 4 (pato)fyziologických relevantních pH podmínkách. Ke stanovení míry chelatace byla použita ferozinová spektrofotometrická metoda. Měření ukázalo závislost chelatačního účinku na pH. Se snižujícím se pH, chelatační účinek klesal. Při pH 7.5 prokázala většina látek 100% chelataci železnatých iontu ve stechiometrickém poměru 1:1. Imbalance of iron levels can lead to serious diseases. Since there is no excretory mechanism, the homeostasis is regulated at the level of absorption in the intestine. The iron overload, which leads to tissue damage due to catalysis of the formation of free radicals, occur because of genetic disorders such as hemochromatosis or owing to frequent administration of transfusions. Rational therapy for iron overload is the administration of the chelators. The aim of this study was to evaluate the ability of derivatives of 2,6,7- trihydroxyxanthen-3-one (synthesized at the University of Sarajevo - Dr. Durić) to chelate iron in 4 (patho) physiologically relevant pH conditions. The ferrozine spectrophotometric method was used to determine the degree of chelation. Measurements showed dependency of the chelating effect on pH: lowering pH resulted in the decrease of the effect.
