Nádorová onemocnění patří mezi jeden z nejzávažnějších problémů vyspělé civilizace a současné medicíny. Neuroblastom je dětský solidní nádor embryonálního typu vyvíjející se z buněk neurální lišty. Významnou roli v patogenezi tohoto onemocnění hraje zvýšená exprese některých onkogenů a jejich mutace vedoucí k poruše regulací. Jednou z možností léčby nádorů je využití cytostatik, málo selektivních látek, které svým toxickým účinkem usmrcují jak nádorové, tak zdravé buňky. Příkladem takové cytotoxické látky je cisplatina (CDDP) vázající se na purinové báze a vedoucí buňku k apoptose. Častou komplikací léčby je vznik rezistence k použitému léku. Ve spojení s touto problematikou je zkoumána vakuolární H+ -ATPasa (V- ATPasa), která je zodpovědná za acidifikaci vakuol. Tato bakalářská práce se zabývá sledováním exprese vybraných genů V-ATPasy v neuroblastomové buněčné linii senzitivní (UKF-NB-4) a rezistentní k cisplatině (UKF-NB-4CDDP ) a po jejich ovlivnění cisplatinou. To bylo provedeno na genové úrovni izolací buněčné RNA, reverzní transkripcí za vzniku komplementární DNA (cDNA) a metodou kvantitativní polymerázové řetězové reakce v reálném čase (RT-PCR). Snahou práce bylo poukázat na možný vliv V-ATPasy na vzniku chemorezistence k cisplatině, a tím přispět k úvahám o V-ATPase jako o možném... Tumour diseases belong among some of the most severe problems of advanced civilization and of current medicine. Neuroblastoma is a solid embryonal children's tumour, which develops from the cells of the neural crest. The increased expression of some oncogenes and their mutation leading to dysfunction plays a major role in the pathogenesis of this disease. One of the possible treatments of the tumours is the use of cytostatics, substances with no selectivity, which kills both damaged and healthy cells by their toxic effect. An example of such a cytotoxic substance is cisplatin (CDDP) which binds with purine nucleotides and leads the cell to apoptosis. A current complication in the treatment is development of chemoresistance. In connection with this issue vacuolar-type H+ -ATPase (V-ATPase) responsible for the acidification of vacuoles is studied. This bachelor thesis concentrates on studying the expression of selected genes of the V- ATPase in the neuroblastoma stem cell line sensitive (UKF-NB-4) and resistant to cisplatin (UKF-NB-4CDDP) and after their exposure to cisplatin.
