Hlavním cílem práce je zhodnotit míru zastoupení jednotlivých typů variantních a slabých RhD antigenů u prvodárců krve, pacientů a těhotných žen. Testování probíhalo na Transfuzním oddělení Fakultní nemocnice Hradec Králové v období od října 2015 do února 2019. K RHD genotypizaci byla použita metoda PCR-SSP pomocí komerčně dodávaných BAGene SSP kitů od firmy BAGene Health Care. Do studie bylo zahrnuto 32 vzorků prvodárců krve ze sledovaného období s cílem určit konkrétní typ RhD antigenu a 188 vzorků pacientů a těhotných žen, u kterých nebylo možné využitím sérologických metod jednoznačně určit RhD antigen. U všech těhotných žen bylo navíc na základě výsledku genotypizace rozhodnuto o nutnosti podání anti-D imunoglobulinu. Provedená RHD genotypizace u všech sérologicky nejasných vzorků umožnila určení konkrétního typu variantního nebo slabého RhD antigenu. V souboru dat prvodárců krve byl slabý RhD antigen detekován v 1,12 % případů z celkového počtu vyšetřených prvodárců krve. V souboru pacientů... Author: Bc. The main aim is to evaluate the rate of the representation of individual types of variant and weak RhD antigens in first-time blood donors, patients and pregnant women. Testing took place at the Transfusion Department of University Hospital Hradec Králové between October 2015 and February 2019. The PCR-SSP method was used for RHD genotyping using commercially supplied BAGene SSP kits from BAGene Health Care. The study includes 32 samples from first-time blood donors in the reference period to determine the specific type of RhD antigen, and 188 samples from patients and pregnant women, for whom serological methods could not be used to unequivocally identify the RhD antigen. For all pregnant women, moreover, the genotyping result was a factor in determining whether to administer anti-D immunoglobulin. This RHD genotyping for all serologically ambiguous samples has made it possible to determine a specific type of partial or weak RhD antigen.
