[cze][eng] Viry Zika a Dengue, náležející do rodiny Flaviridae, způsobují horečnatá onemocnění. Infekce může být bezpříznaková či s příznaky lehké virózy, nicméně se může rozvinout až po život ohrožující komplikace. V případě Dengue může nemoc přerůst až do hemorrhagické horečky. Infekce virem Zika je také nebezpečná pro těhotné ženy kvůli zvýšenému riziku výskytu mikrocefalie u novorozenců. Tyto viry jsou přenášeny komáry rodu Aedes, řadí se tak mezi arboviry. Pro boj s těmito chorobami je vhodná vakcinace, zároveň ale probíhá vývoj virostatik. Inhibitory virových proteas jsou důležitými klinicky schválenými virostatiky proti HCV a HIV. Zda by vhodným cílem nových léčiv mohly být i proteasy virů Dengue nebo Zika není zatím zcela zřejmé, nicméně tyto proteasy jsou velmi zajímavé i z pohledu základního výzkumu. Flavivirová proteasa se nachází v polyproteinu syntetizovaném aparátem hostitelské buňky, a to v doméně NS3. Tento enzym potřebuje aktivační doménu NS2B, mechanismus interakce však není zcela prostudován. NS3 je multidoménový enzym, kromě proteasové aktivity vykazuje ještě aktivitu NTPasovou, helikasovou a RNA 5'-trifosfatasovou. Není zřejmé, proč tyto enzymy koexistují v jedné proteinové doméně. Abychom mohli odpovědět na některé tyto otázky, je vhodné připravit expresní systémy pro zkoumání enzymové...in English Zika and Dengue viruses, which are part of Flaviridae family, are responsible for fever diseases. Infection can be asymptomatic or with symptoms of light virosis, however, it could be also life threatening. In the case of Dengue, it could progress to haemorrhagic fever. Infection by Zika is also dangerous for pregnant women because of enhanced risk of microcephaly in new-borns. These viruses are transferred by mosquitoes from Aedes family and are part of arbovirus group. Vaccination is a good tool for control of these diseases, but development of virostatics are also in progress. Inhibitors of viral proteases are important clinically approved virostatics against HIV and HCV. It is not clear, if Zika of Dengue proteases can become drug targets, however these proteases are also interesting for basic research. Flavivirus protease is located in viral polyprotein synthesized by host cells as a part of NS3 domain. This enzyme needs activation domain NS2B; mechanism of this interaction is not fully understood. On top of the protease activity, NS3 domain includes helicase, NTPase and RNA 5'-triphosphatase activity. It is not known why these enzymes coexist as one protein domain.
