Suprachiasmatická jádra (SCN) jsou řídící centrum cirkadiánních rytmů savců. Pro udržení přibližné 24hodinové periody svých rytmů vyžadují synchronizaci s fází vnějšího prostředí. Nejvýznamnějším vnějším synchronizátorem určujícím periodu rytmů v SCN je pravidelné střídání světla a tmy. Světelná informace je převáděna ze sítnice do ventrolaterální části SCN, kde zásadní úlohu při přenosu světelného signálu mají na excitačních synapsích přítomné ionotropní glutamátové receptory (iGluRs). Posttrasnkripční úpravy RNA mohou měnit funkční vlastnosti iGluRs a přispívat tak k synaptické plasticitě. Míra těchto posttranskripčních úprav může být in vitro ovlivněna farmakologickou manipulací neuronální aktivity. Cílem této práce bylo určit, do jaké míry dochází k posttranskripčním úpravám iGluRs v in vivo modelu potkaního SCN, a dále čím jsou tyto úpravy regulovány. Metodou PCR a následného sekvenování amplifikované DNA jsme analyzovali posttranskripční úpravy RNA u GluA2 podjednotky AMPA receptoru (AMPAR) a GluK2 podjednotky kainátového receptoru. Kromě toho jsme metodou kvantitativního PCR stanovili expresi mRNA pro GluA1 a GluA2 podjednotku AMPAR a editačního enzymu ADAR2 v SCN. Výsledky ukázaly, že míra RNA editace se u sledovaných podjednotek iGluRs zvyšuje v podmínkách světlem indukované neuronální... Suprachiasmatic nuclei (SCN) are primary center of mammalian circadian rhythms. To maintain a 24 hour period of its rhythms, SCN are synchronized with phase of external environment. Regular changes of light and darkness are known to be the main external synchronizer that determines the period of SCN rhythms. Information about light is being transferred from retina to the ventrolateral region of SCN through excitatory synapses where ionotropic glutamate receptors (iGluRs) play a primary role in the signal transduction. Posttranscriptional modifications of RNA can alter the functional properties of iGluRs, thus this process contributes to synaptic plasticity. The extent of posttranscriptional modifications of iGluRs can be in vitro affected by neuronal activity altered by pharmacological manipulation. The aim of this study was to determine possible changes of posttranscriptional modifications of iGluRs in in vivo rat SCN model and how this process can be regulated. RNA posttranscriptional modifications of GluA2 subunit of AMPA receptor (AMPAR) and GluK2 subunit of kainate receptor were assessed using PCR and subsequent sequencing of amplified DNA.
