Hadi (Serpentes) jsou skupinou šupinatých plazů (Squamata) čítající více než 3600 druhů. Většina hadů patří do skupiny Caenophidia, ta tvoří asi 90 % všech moderních hadů a je také nejstudovanější hadí skupinou. Přestože jsou šupinatí plazi v determinaci pohlaví a uspořádání genomu variabilní, přinejmenším hadi skupiny Caenophidia jsou v tomto ohledu skupinou velice stabilní. Typický - a zřejmě také ancestrální - karyotyp hadů obsahuje 36 chromozomů s 16 makro- a 20 mikrochromozomy. U všech druhů hadů se předpokládá genotypově určené pohlaví (GSD) a až do nedávna se mělo za to, že všichni hadi mají ZZ/ZW pohlavní determinační systém. Hlavní důvod této domněnky spočívá v tom, že byli studováni převážně hadi skupiny Caenophidia a u zbývajících linií se jen předpokládalo, že mají homologické pohlavní chromozomy, ačkoli se u těchto hadů - s jedinou výjimkou (Acrantophis dumerili) - pohlavní chromozomy popsat nepodařilo. Teprve současná studie ukázala, že minimálně u krajt a hroznýšů došlo k nezávislému vzniku pohlavních chromozomů XX/XY. Pohlavní chromozomy těchto hadů jsou však homomorfní a klasickými cytogenetickými metodami se je prozatím nepodařilo odhalit. Proto bylo jedním z cílů této práce zjistit, zda je možné identifikovat slabě diferencované pohlavní chromozomy u hadů mimo skupinu Caenophidia... Snakes (Serpentes) are a group of squamate reptiles (Squamata) including more than 3600 species. The vast majority of snakes are from the group Caenophidia, which includes approximately 90% of all extant snake species and represents the most studied lineage. Squamate reptiles are variable in sex determination and genome organisation, however, caenophidian snakes are quite stable in these respects. The typical - and probably ancestral - snake karyotype is composed of 36 chromosomes with 16 macro- and 20 microchromosomes. In all snake species, genotypic sex determination (GSD) is expected and it was assumed until recently that all snakes possessed ZZ/ZW sex chromosomes. The main reason for this is that most of the studied snakes were from the group Caenophidia and in the rest of the snake lineages it was believed that their sex chromosomes were homologous. In fact, the sex chromosomes of non-caenophidian snakes have not yet been identified - with the single exception of Acrantophis dumerili. Nevertheless, a recent study showed that there was an independent evolution of XX/XY sex chromosomes in pythons and boas.
