Cystická fibrosa je autozomálně recesivní onemocnění způsobené mutací genu CFTR, která vede k poškození stejnojmenného proteinu s funkcí iontového kanálu. V důsledku poškození dochází k narušení rovnováhy elektrolytů. Nejvíce je postiženo dýchací ústrojí, kde dochází k zahuštění hlenu a ke změnám v glykosylaci povrchových struktur buněk plicního epitelu. Výše uvedené změny vedou k nedostatečné obranné funkci plic a větší náchylnosti k bakteriálním infekcím. Nejčastějším patogenem dýchacích cest pacientů s CF je Pseudomonas aeruginosa. Tato bakterie obsahuje řadu virulentních faktorů a má schopnost tvořit biofilm, který ji chrání před obrannými mechanismy hostitele a účinky antibiotik. Léčba antibiotiky je navíc komplikovaná vznikem rezistencí. Slepičí protilátky vykazují značný potenciál jako prostředek prevence bakteriálních infekcí plic u pacientů s CF. Efekt specifických a nespecifických protilátek na adherenci kontrolního kmenu Pseudomonas aeruginosa (PAK, ST 1763) ex vivo byl sledován na modelovém systému buněčných linií plicních epiteliálních buněk od pacienta s CF (CuFi-1) a zdravého jedince (NuLi-1). Bakteriální a plicní buňky byly značeny fluorescenčními barvivy PKH, vyhodnocení bylo prováděno spektrofluorimetricky. Bylo ověřeno, že slepičí protilátky proti PA-IIL mají schopnost specificky... Cystic fibrosis is an autosomal recessive disease caused by mutation of the CFTR gene. This mutation results in the damage of protein with ion channel function. As a consequence of this damage, electrolyte equilibrium is impaired. The respiratory tract is affected the most because of the mucus thickening and changes in the glycosylation of lung epithelium cell surface structures. These changes lead to an insufficient defense function of lungs and greater susceptibility to bacterial infections. The most common pathogen of respiratory tract of CF patients is Pseudomonas aeruginosa. This bacterium contains numerous virulence factors and has the ability to form a biofilm that protects it against the host defense mechanisms and antibiotic effect. Antibiotic treatment is also complicated by the development of bacterium resistance. Chicken antibodies have a considerable potential as a tool for preventing bacterial lung infections of CF patients. The influence of specific and nonspecific antibodies on the adherence of Pseudomonas aeruginosa (PAK, ST 1763) ex vivo was monitored on a model system of lung epithelia cells from a CF patient (CuFi-1) and a healthy individual (NuLi-1).
