Histonové modifikace ovlivňují celou řadu buněčných dějů, jedním z nich je i oprava poškození DNA. Tato práce se zaměřuje na metylaci lysinu 36 histonu 3 (H3K36). V první části práce je popsána role této modifikace v lokalizaci dvouvláknových zlomů DNA v germinálních buňkách. Dvouvláknové zlomy zde iniciují meiotickou rekombinaci, která je nezbytná pro správný průběh meiózy. V této práci jsou podrobněji popsány celkem tři metyltransferázy histonů. První z nich je PRDM9, enzym exprimovaný pouze v oocytech a spermatocytech během meiotické profáze a zodpovědný za rozmístění hotspotů rekombinace u většiny savců. Druhá část práce je zaměřena na roli metylace H3K36 na opravy poškození DNA v somatických buňkách pomocí homologní rekombinace (HR) a nehomologního spojení konců DNA (NHEJ). V druhé části práce jsou popsány metyltransferázy SETD2 a SETMAR. SETD2 trimetyluje H3K36 a H3K36me3 je rozeznávána proteinem LEDGF, díky kterému jsou k DNA naváděny další komponenty nutné pro HR. Protein SETMAR dimetyluje H3K36 a spolu s touto histonovou modifikaci podporuje opravu DNA zlomů pomocí NHEJ. Studium metylace H3K36 je důležité pro lepší pochopení jednotlivých způsobů oprav DNA. Správná oprava DNA zlomů je nutná pro udržení celistvosti genomu a potlačení vzniku rakoviny. Chybná regulace metylace H3K36 proto... Histone modifications affect many cellular processes including DNA damage repair. This thesis focusses on the methylation of lysine 36 of histone 3 (H3K36). The role of this modification in the localization of DNA double-stranded breaks in germinal cells is described in the first part of this thesis. Double-stranded breaks initiate meiotic recombination, which is essential for successful meiosis. This thesis also describes three histone methyltransferases. The first is PRDM9, an enzyme expressed only in oocytes and spermatocytes during meiotic prophase and responsible for the localization of recombination hotspots in most mammals. The second part of this thesis deals with the role of H3K36 methylation in DNA damage repair in somatic cells using homologous recombination (HR) and nonhomologous DNA ending joining (NHEJ). The proteins SETD2 and SETMAR are described in the second part. SETD2 trimethylates H3K36, and H3K36me3 is recognized by the LEDGF protein. Through LEDGF, other components necessary for HR are recruited to DNA. SETMAR dimethylates H3K36 and together with this histone modification promote DNA break repair with NHEJ.
