1. SOUHRN Regenerace jater po částečné hepatektomii u potkanů s nealkoholovým ztukovatěním jater Nealkoholové ztukovatění jater (NAFLD - non-alcoholic fatty liver disease) je v současnosti nejčastějším chronickým onemocněním jater v ekonomicky vyspělých zemích s prevalencí kolem 30 %. Resekce jater z různých příčin je nyní běžnou součástí klinické praxe. Úspěšné zotavení a obnova jaterních funkcí závisí na regenerační schopnosti zbytku jater. Průběh regenerace jater může být velmi významně ovlivněn současným postižením jater patologickým procesem včetně NAFLD. Cílem této práce bylo studium časné fáze regenerace jater po částečné hepatektomii (PHx) u potkanů s nutričně navozenou prostou steatózou. Nejprve jsme zavedli model NAFLD podáváním diety s vysokým obsahem tuků po dobu 3 resp. 6 týdnů potkanům kmene Wistar a Sprague-Dawley. U obou kmenů došlo k rozvoji steatózy bez známek zánětu, nekrózy nebo fibrotických změn. V druhé části naší studie jsme se sledovali, zda regenerace jater po PHx je u potkanů s NAFLD alterována. Regeneraci jater jsme posuzovali především ze změn inkorporace bromdeoxyuridinu do DNA hepatocytů. Regenerace jater postižených prostou steatózou navozená 2/3 PHx není signifikantně ovlivněna v porovnání s neztukovatělými játry. Jedinou změnou v regenerační odpovědi, kterou jsme pozorovali... 1. SUMMARY Liver regeneration after partial hepatectomy in rats suffering from non-alcoholic fatty liver disease Non-alcoholic fatty liver disease (NAFLD) is currently the most frequent chronic liver disease in economically developed countries with prevalence of about 30 %. Liver resection from different reasons is nowadays common surgical procedure. Successful recovery and renewal of liver functions depend on regenerative capacity of liver remnant. The course of liver regeneration could be profoundly influenced by concomitant liver pathological processes including NAFLD. The aim of this work has been study of early phase of liver regeneration after partial hepatectomy (PHx) in rats with nutritionally induced simple steatosis. At the beginning we introduced a model of NAFLD by a high-fat diet giving to Wistar or Sprague-Dawley rats for 3 and 6 weeks, resp. This regimen induced simple steatosis without signs of inflammation, necrosis or fibrotic changes in both strains. In the second part of our study we followed if liver regeneration after PHx in rats with NAFLD is altered. Liver regeneration was assessed by bromodeoxyuridin incorporation into DNA of hepatocytes.
