Protein-protein interactions play an important role in nearly all processes of the living cells and the function of many proteins is dependent on their specific interactions with other biomolecules. A reliable tool to modulate these interactions would be invaluable for the development of molecules suitable for diagnostics, medicine, and biotechnology. In this work, we aimed to study the specificity of interactions in the model system of Interferon gamma receptor 1 (IFNgR1) and its natural ligand Interferon gamma (IFNg), important in innate immunity. We searched for mutations within the interferon receptor molecule IFNgR1 to modulate (increase as well as decrease) its affinity to IFNg by in silico analysis of the existing crystal structures of the complex between IFNgR1 and IFNg. We modeled amino acid substitutions and gauged how they influenced the interaction using empirical force field implemented in software FoldX. All selected promising IFNgR1 variants were expressed in Escherichia coli, purified to homogeneity, characterized, and kinetics of their interactions with IFNg was measured by Surface Plasmon Resonance (SPR). Téměř ve všech dějích živých buněk hraje interakce mezi proteiny důležitou roli a funkce mnohých proteinů je závislá na jejich specifické interakci s ostatními biomolekulami. Pro vývoj molekul vhodných pro diagnostiku, medicínu a biotechnologie by bylo velmi přínosné, kdybychom měli k dispozici nástroje k ovlivňování interakcí mezi proteiny. Tato práce se zabývala specificitou interakcí mezi proteiny na modelovém příkladu receptoru 1 pro interferon gama (IFNgR1) a jeho přirozeném vazebném ligandu interferonu gama (IFNg), jehož funkce je důležitá pro přirozenou imunitu. Při hledání mutací proteinu IFNgR1, které by měly měnit jeho vazbu (zvýšit nebo snížit) k IFNg, byla použita počítačová in silico analýza dostupných krystalových struktur komplexu mezi IFNgR1 a IFNg. Záměny aminokyselin byly počítačově modelovány a jejich vliv na interakci byl počítán programem FoldX. Všechny varianty receptoru 1, které vyhovovaly kritériím výběru, byly produkovány v bakterii Escherichia coli, dále purifikovány, charakterizovány a jejich interakce byla změřena na přístroji SPR, jenž měří povrchovou rezonanci plazmonů (Surface Plasmon Resonance). První skupina variant IFNgR1 obsahovala mutace na interakčním rozhraní s IFNg. Z SPR měření vyplývá, že většina těchto variant vykazovala stejnou vazbu k IFNg jako nezměněný...
