[cze][eng] Cystická fibróza je autozomálně recesivní onemocnění, které patří k nejčastějším dědičným poruchám. Tato nemoc je způsobena mutací v genu CFTR kódujícím chloridový kanál, čímž dochází k poruše transportu iontů a výraznému zvýšení viskozity hlenů. Nejvíce postižen je respirační systém. Zvýšení obsahu solí a přítomnost hustého hlenu v plicích potlačují antibakteriální účinky imunitního systému. Bakteriální infekce jsou nejčastější příčinou smrti pacientů s CF, zejména infekce Pseudomonas aeruginosa. Nejčastěji je pro terapii infekce tímto patogenem používána léčba antibiotiky, která je komplikována častým vznikem rezistencí. Jsou proto hledány nové možnosti terapie. Nadějnou by mohla být pasivní imunizace pacientů pomocí žloutkových protilátek IgY. Pro sledování vlivu těchto protilátek in vivo byl zvolen modelový systém buněk plicního epitelu zdravého pacienta (NuLi-1), pacienta trpícího CF (CuFi-1) a 3 kmenů bakterií Pseudomonas aeruginosa. Byly porovnávány protilátky proti lektinu Pseudomonas aeruginosa PA-IIL, který je významnou adhezivní strukturou této bakterie, pocházející od 2 různých imunizovaných zvířat. K vizualizaci a vyhodnocení adheze bakterií na plicní buňky v přítomnosti protilátek bylo používáno barvení fluorescenčními sloučeninami PKH a následné spektrofluorimetrické vyhodnocení....A b stra ct C ystic fibrosis is an autosom alrecessive disease that is one of the m ost com m on hereditary disorders. T he disease is caused by the m utation in the gene encoding C F T R chloride channe l w hich leads to the failure of ion transportand a significant increase in viscosity o f m ucus,affecting especially the respiratory system .T he increase of saltand the presence of thick m ucus in the lungs suppress the antibacterialeffects of the im m une system .B acterial infections are the m ost com m on cause of death in patients w ith C F, especially the P seudom onas aerugino sa infection. A ntibiotics are used for treatm ent of the infection by this pathogen,butthe treatm ent is frequently com plicated by developing resistance.D ue to this fact, new w ays of treatm ent have been searched for. T he m ethod of passive im m unization of patients w ith yolk IgY antibodies seem s to be prom ising.
