[cze][eng] Hlavní cirkadiánní oscilátory savců, suprachiasmatická jádra (SCN), jsou uložena v hypotalamu podél třetí mozkové komory. Jsou zodpovědná za časovou synchronizaci behaviorálních a fyziologických procesů v organismu. Jejich endogenní perioda a fáze jsou nastavovány rytmickými změnami ve vnějším prostředí, zejména střídáním světla a tmy. Světelný puls v době noci aktivuje glutamátové receptory typu NMDA, neuronální NO syntázu a MAPK kinázu ERK1/2. Tato signalizační dráha pak ovlivňuje transkripci hodinových genů a tím celý mechanismus generování cirkadiánních oscilací. Protein Dexras1 působí jako nukleotidový exchanger monomerního i heterotrimerního Gi/o proteinu a je považován za mezičlánek mezi aktivací glutamátových receptorů agonistou NMDA, nNOS signalizací a forsforylací ERK1/2. Je rytmicky exprimován v suprachiasmatickém jádře a jeho genetická delece vede k abnormální světelné synchronizaci. To může naznačovat jeho úlohu v signální kaskádě, která vede informaci o světelném pulzu z retiny do SCN. Cílem diplomové práce bylo popsat expresi mRNA a proteinu Dexras1 ve vývoji suprachiasmatických jader potkana a korelovat jí s maturací mechanismu světelné synchronizace. Naše výsledky ukazují, že Dexras1 je rytmicky exprimován s nízkou amplitudou již u 3 -denních mláďat, avšak lokalizace jeho exprese...The main mammalian circadian oscillators, suprachiasmatic nuclei (SCN), are stored in the hypothalamus along the third ventricle. They are responsible for time synchronization of behavioral and physiological processes in the body. The endogenous period and phase are controlled by rhythmic changes in the external environment, especially by changing of light and darkness. The light pulse during the night activates NMDA type glutamate receptors, neuronal NO synthase kinase and MAPK ERK1 / 2. This signaling pathway affects the transcription of clock genes and thus the mechanism generating circadian oscillations. Protein Dexras1 acts as a nucleotide exchanger monomer and heterotrimer Gi/o protein and is considered a connecting link between the activation of NMDA glutamate receptor agonist, nNOS signaling and phosforylation of ERK1 / 2. It is rhythmically expressed in the suprachiasmatic nucleus and its genetic deletion leads to abnormal light synchronization.This may indicate its role in the signaling cascade that results in the information concerning the light pulse from the retina to the SCN.
