[cze][eng] Izolovaný deficit F1FO-ATPsyntázy je skupinou mitochondriálních onemocnění způsobených mutacemi v jaderně- a mitochondriálně-kódovaných strukturních podjednotkách, nebo jaderně-kódovaných faktorech biogenese komplexu F1FO-ATPsyntázy. Nejčastěji jsou to mutace v genu MTATP6, nacházející se v mitochondriální DNA, a v genu TMEM70, nacházející se v jaderné DNA. Gen MTATP6 kóduje strukturní podjednotku "a" komplexu F1FO-ATPsyntázy a jeho mutace způsobuje snížené množství F1FO-ATPsyntázy a její fosforylační aktivitu. Gen TMEM70 kóduje protein vnitřní mitochondriální membrány o velikosti 21 kDa s doposud zcela neobjasněnou funkcí a jeho mutace způsobuje poruchu biogeneze komplexu F1FO-ATPsyntázy. Cílem práce bylo studovat ve dvou liniích primárních kožních fibroblastů s izolovaným deficitem F1FO-ATPsyntázy (mutace m.8851T>C v MTATP6 a mutace c.317-2A>G v TMEM70) dopad tohoto deficitu na systém oxidační fosforylace, vybrané mitochondriální proteiny a mitochondriální síť ve srovnání s kontrolní linií primárních kožních fibroblastů v prvních dnech kultivace (1-5 dní) v médiu obsahujícím jako zdroj sacharidů galaktózu nebo glukózu a v médiu s přítomnou a nepřítomnou aminokyselinou L-glutaminu. U kontrolní linie kultivované v médiu DMEM obsahujícím jako zdroj sacharidů galaktózu bylo nalezeno vyšší množství...Isolated deficiency of F1FO-ATPsynthase is a soubgroup of mitochondrial diseases caused by mutations in nuclear and mitochondrial-encoded structural subunits, or nuclear-encoded assembly factors of F1FO-ATPsynthase. The most often mutations are found in a MTATP6 gene localized in the mitochondrial DNA and a TMEM70 gene, localized in the nuclear DNA. A MTATP6 gene encodes subunit a of F1FO-ATPsynthase and its mutation usually leads to reduced phosphorylation activity of F1FO-ATPsynthase. A TMEM70 gene encodes a 21 kDa mitochondrial protein of the inner mitochondrial membrane of not completely explained function and its mutation results in the decrease in a content of fully assembled F1FO- ATPsynthase. The aim of this thesis was to investigate the impact of isolated F1FO- ATPsynthase deficiency on the oxidative phosphorylation system (complex I-IV), other selected mitochondrial proteins, and mitochondrial network in two cell lines of primary human skin fibroblasts with an isolated deficiency of F1FO-ATPsynthase (mutation m.8851T>C in MTATP6 and mutation c.317-2A>G in TMEM70) during the first days of their cultivation in media containing galactose or glucose as a carbohydrate source with a presence or absence of L-glutamine.
