Během nepříznivých podmínek je v eukaryotických buňkách inhibována translace některých mRNA a přednostně jsou syntetizovány proteiny, které se podílejí na boji buňky se stresem. Takto ušetřená energie je využita na opravy poškození buněčných struktur. Nepřekládané mRNA se v podobě ribonukleoproteinových komplexů hromadí a důsledkem je vznik cytoplazmatických stresových granulí. V nich dochází k úpravě struktury a složení těchto komplexů a následně je rozhodováno o jejich osudu - mohou být uskladněny, degradovány nebo vráceny do cytoplazmy pro znovuzahájení jejich překladu. Molekuly mRNA s sebou do stresových granulí přinášejí i proteiny, které se na ně vážou. Mezi takovými proteiny mohou být i aktéři signálních drah a stresové granule se tak nepřímo podílejí na regulaci buněčných dějů, jako je například apoptóza, a hrají tak důležitou roli v přežívání buňky během nepříznivých podmínek. Tato práce pojednává o proteinovém složení stresových granulí v lidských buněčných liniích, stručně charakterizuje stresové faktory, které ke vzniku stresových granulí vedou a věnuje se rozdílům ve složení stresových granulí indukovaných různými stresovými faktory. Součástí práce je tabulka shrnující proteiny, které se v těchto granulích vyskytují. Ve druhé části se práce věnuje charakterizaci proteinů z rodiny... During unfavourable conditions eukaryotic cells inhibit translation of certain mRNAs and preferably synthesize proteins that are involved in the stress response. The saved energy is used for repair of cellular damages. The untranslated mRNAs are accumulated in the form of ribonucleoprotein complexes. This accumulation results in the formation of the cytoplasmic stress granules. These granules are sites of structure remodeling and triage of the ribonucleoprotein complexes - they can be stored, degraded or sent back to the cytoplasm for translation reinitiation. The mRNA molecules carry their associated proteins, which include also proteins implicated in the cell signaling. Stress granules can thus indirectly regulate some processes, such as apoptosis, and play role in the survival of the cell. This thesis focuses on protein content of stress granules in human cell lines, briefly characterizes stress factors that induce their formation and discusses differences between the content of stress granules induced by different stress stimuli. An important part of this thesis is a table summarizing proteins found in the stress granules.
