Alternativním sestřihem mRNA může vznikat z jediného eukaryotického genu několik různých proteinů s odlišnou sekvencí aminokyselin, strukturou, a tedy i funkcí. Tento jev je složitě regulován. Může hrát významnou roli v nádorovém bujení. Například u alternativního sestřihu osteopontinu byl prokázán jeho vliv na metastazování a progresi nádoru prsu. Glutamátkarboxypeptidasa II (též prostatický specifický membránový antigen) se vyskytuje ve zvýšeném množství ve zdravé i nádorové prostatické tkáni. Je známo několik jejích sestřihových variant. U některých z nich byla zkoumána souvislost mezi jejich přítomností a postupem karcinomu prostaty v pacientech pomocí kvantitativního RT-PCR. O expresi dalších sestřihových variant nejsou zatím k disposici žádné přesnější údaje. Mezi ně patří varianty s vynechaným exonem 6 anebo exonem 18. Cílem této práce bylo určit množství právě těchto delecí v několika biologických vzorcích. Za pomoci kvantitativní polymerasové řetězové reakce bylo stanoveno množství delecí exonu 6 a exonu 18 v cDNA kódující prostatický specifický membránový antigen ze vzorků tkání pacientů s benigní prostatickou hyperplazií, nádorem prostaty a z buněčné linie odvozené od karcinomu prostaty. Zároveň bylo stanoveno množství delecí v jednotlivých sestřihových variantách tohoto proteinu, které... Alternative splicing is a mechanism of generating distinct proteins that are encoded by the same gene. These proteins differ in amino acid sequence, overall structure and function. Splicing dysregulations have been shown to be implicated in several pathologic processes including cancer. For example, non-physiological splicing of osteopontin was proved to play a key role in cell progression of breast cancer. Glutamate carboxypeptidase II (also called prostate specific membrane antigen, PSMA) is present in both normal prostate and prostate cancer. Several splice variants of PSMA have been described and it has been suggested that the overexpression of some of them could be involved in the progression of prostate cancer. Nevertheless, more detailed investigation of each of the PSMA splice variant in terms of their occurrence in prostate cancer cells remains to be performed. This thesis focuses on the exploration of the expression of PSMA splice variants with deleted exons 6 and 18 in samples of a cell line derived from human prostate cancer, benign prostate hyperplasia and prostate cancer.
