Úvod: Molekuly hlavního histokompatibilního komplexu (MHC) hrají důležitou úlohu v regulaci imunitní odpovědi a udržování imunitní homeostázy. Regulace jejich exprese je proto klíčovým faktorem ovlivňujícím adaptivní imunitní odpověď. Jedním z mechanismů této regulace je methylace DNA v promotorových oblastech, která ovlivňuje dostupnost DNA pro transkripční faktory. Stárnutí organismu je spojeno se změnami v methylaci DNA a zvýšená predispozice k rozvoji autoimunitních onemocnění ve stáří by proto mohla být spojena právě se změnami v methylačním stavu regulačních oblastí genů MHC II. třídy. Cíle: Cílem této práce je analýza methylačního stavu regulačních oblastí genů DQA1 a DQB1 a porovnání rozdílů mezi generacemi a jednotlivými alelami. Dále je porovnána úroveň exprese mRNA genu DQA1 mezi generacemi a mezi jednotlivými alelami. Metody: Z krve odebrané dárcům ze tří různých věkových skupin byly izolovány DNA a RNA. DNA byla modifikována bisulfitovou konverzí, dále byly amplifikovány regulační oblasti genů DQA1 a DQB1 a tyto úseky byly klonovány do bakterií. Pozitivní klony byly sekvenovány a podrobeny methylační analýze. RNA byla pomocí reverzní transkripce přepsána do cDNA a relativní úroveň její exprese byla poté určena pomocí kvantitativní PCR. Background: The major histocompatibility complex (MHC) molecules play an important role in the immune response regulation and in the maintenance of the immune homeostasis. Regulation of their expression is therefore a key factor influencing the adaptive immune response. DNA methylation of gene regulatory regions is one of the mechanisms of gene expression control that affects the accessibility of DNA to transcription factors. Ageing is connected with changes in DNA methylation and increased predisposition to autoimmune diseases in older age could be associated with changes in MHC class II genes methylation. Aims: The aim of this diploma thesis is to analyze the methylation profile of DQA1 and DQB1 genes regulatory regions and to compare its differences between the generations and between individual alleles. The next aim is to compare DQA1 mRNA expression between the generations and between single alleles. Methods: DNA and RNA were isolated from blood of three age group donors. DNA was converted by the bisulfite treatment and regulatory regions of HLA class II genes were amplified and cloned into bacteria. Positive clones were sequenced and then analyzed.
