[cze][eng] Perfluorocarbony (PFC) jsou uhlovodíky, ve kterých jsou všechny nebo některé z atomů vodíku nahrazeny atomy fluoru. PFC mají velmi vysokou kapacitu pro rozpoustnost kyslíku. Jsou chemicky a biologicky inertní. Nejúspěšnější klinická aplikace PFC je tzv. "two-layer method" (TLM) určená pro konzervaci pankreatu před izolací Langerhansových ostrůvků (LO). TLM je metoda konzervace založená na kombinaci okysličeného PFC a UW roztoku, kterým je PFC převrstvený. V experimentu byla TLM úspěšně použita při transplantaci srdce a střeva. V našich experimentech na syngenních potkanech jsme testovali vliv dlouhodobé studené konzervace pomocí perfluorocarbonu na prevenci tkáňového poškození a zlepšení výsledků transplantace ledviny, pankreatu a LO. U transplantace ledviny a LO jsme použili jako metodu konzervace TLM. V případě transplantace pankreatu jsme pracovali s pefluorohexyloctanem patřícím do nové generace méně lipofilních PFC. 1. Ledviny byly konzervovány po dobu 24 hodin buď v UW roztoku (n = 16), pomocí TLM (n = 16), nebo byly transplantovány ihned (kontrolní skupina, n = 12). U poloviny zvířat z každé skupiny bylo hodnoceno přežívání, u ostatních byly odebrány štěpy k semikvantitativnímu histologickému hodnocení a měření výskytu apoptózy 24 hodin po transplantaci. Dlouhodobé přežívání v UW, TLM a...(English) Perfluorocarbons (PFC) are hydrocarbons in which some or all of the hydrogen atoms are replaced with fluorine. PFC have a very high capacity for dissolving oxygen. They are chemically and biologically inert. The most successful clinical application of PFC is the "two-layer method" for pancreas preservation before islet isolation. The two-layer organ preservation method (TLM) is based on oxygenated perfluorocarbon overlaid with University of Wisconsin (UW) solution. In experiment it has been successfully used for heart and intestine transplantation. We tested whether this technique would prevent tissue damage and improve results of kidney, pancreas and islets of Langerhans transplantation with prolonged ischemia time in an experimental model of syngenic rats. In kidney and islets of Langerhanse transplantation model we used TLM preservation method. In pancreas transplantation model we used perfluorohexyloctane (PFH) as a new generation of less lipophilic PFC. 1. Kidneys were stored for 24 hours either in UW solution (n = 16), with TLM (n = 16) or transplanted immediately (control group, n = 12). In half of the animals, survival was observed and in the other animals grafts were procured for semiquantitative histological scoring and TUNEL apoptosis assessment 24 h after transplantation....
