Hlavným cieľom tejto bakalárskej práce je optimalizácia mikroreologických techník pre charakterizáciu biopolymérnych hydrogélov. K tomu boli vyuţité hydrogély na základe termoreverzibilného biopolyméru – agarózy. Skúmaný bol vplyv inkorporovaného polyelektrolytu (polystyrensulfonátu sodného) na štruktúru a viskoelastické vlastnosti agarózového hydrogélu, a to prostredníctvom fluorescenčných značených častíc, ktoré boli začleňované do štruktúry gélu. Zmena výslednej štruktúry bola najprv pozorovaná prostredníctvom difúznych koeficientov a po úspešnej optimalizácii boli stanovené aj MSD krivky. Meranie bolo uskutočnené fluorescenčnou korelačnou spektroskopiou.
The main aim of the bachelor thesis is optimalization of microrheological techniques for characterization of biopolymer hydrogels. Hydrogels, based on thermoreversible biopolymer agarose, were selected for these experiments. The influence of incorporated poly(sodium 4-styrenesulfonate) on structure and viscoelastic properties of agarose hydrogels was investigated through diffusion coefficient and MSD curves. Microrheological properties were studied by fluorescence correlation spectroscopy, by using fluorescent beads, which were embedded in hydrogel systems.
