Kmen Rhodococcus erythropolis A4 je zdrojem enzymů nitrilhydratázy a amidázy katalyzujících konverze nitrilů a amidů. Tyto enzymy jsou používány při průmyslových biotransformacích a bioremediacích. Vzhledem k tomu, že genové manipulace vedoucí ke zvýšení produkce těchto enzymů bylo v kmeni A4 obtížné provést, byly v této práci identifikovány a analyzovány příslušné geny (ami a nha1+nha2) v příbuzném kmeni R. erythropolis CCM2595, v kterém lze manipulace s plazmidy i v chromozomu rutinně provádět. Geny ami a nha1+nha2 z kmene R. erythropolis CCM2595 byly izolovány a společně se sousedícími oblastmi (celkem 5,5 kb) sekvenovány. Byla zjištěna organizace těchto genů a předpokládaných regulačních genů v kmeni CCM2595 a zkoumán způsob regulace exprese těchto genů. Pro analýzu transkripce genů pro amidázu a nitrilhydratázu z obou kmenů R. erythropolis byl použit promoter-probe vektor pEPR1 replikující se v Escherichia coli a R. erythropolis, v němž byly zkonstruovány transkripční fúze promotorů Pami z kmene A4 i CCM2595 a reportérového genu gfp. Aktivita promotorů Pami byla měřena prostřednictvím fluorescence produktu genu gfp, zeleného fluoreskujícího proteinu. Měření fluorescence buněk nesoucích vektor pEPR1 s promotory genů ami z obou zkoumaných kmenů prokázalo, že jejich aktivita je převážně konstitutivní a... The strain Rhodococcus erythropolis A4 is a source of enzymes nitrilhydratase and amidase, that catalyse conversion of nitriles and amides. These enzymes are used in industrial biotransformation and bioremediation. Since it was difficult to carry out genetic manipulations aimed at increasing the production of these enzymes in the strain A4, the corresponding genes (ami and nha1 + nha2) of a related strain R. erythropolis CCM2595, in which both plasmid and chromosome manipulations can be routinely performed, were identified and analyzed in this diploma theses. The ami and nha1 + nha2 genes from the strain R. erythropolis CCM2595 were isolated and sequenced together with the flanking regions (5.5 kb in total). The organization of these genes and the expected regulatory genes was described in the strain CCM2595 and mechanisms of regulation of expression of these genes were studied. For the analysis of transcription of amidase and nitrilhydratase genes from both strains of R. erythropolis, the promoter-probe vector pEPR1 replicating in Escherichia coli and R. erythropolis was used. Transcriptional fusion of Pami promoters of the strains A4 and CCM2595 and the reporter gfp gene were constructed.
