V okolním prost edí bakterií dochází k neustálým zm nám životních podmínek. Jednou z možností jak se bakterie mohou na tyto zm ny adaptovat by mohlo být zvýšení muta ní rychlosti. Mutabilita bakterií je za normálních okolností udržována na velice nízké hladin pomocí r zných kontrolních a opravných mechanism . Jedním z nich je mismatch repair systém, který se výrazn podílí na udržení genetické stability organism . Vy azením funkce tohoto systému, dochází k vzniku v tšího po tu mutací v bu ce. V p ípad , že se bakteriální populace nachází v nep íznivém prost edí, m že být tato vlastnost výhodou v adaptaci na nové životní podmínky. Do jaké míry se tyto mechanismy podílí na adapta ních procesech u Bacillus subtilis zatím není prokázáno. Na adaptivní mutagenezi proto byla zam ena p edchozí práce z naší laborato e, která se zabývala vytvo ením experimentálního systému pro kvantifikaci mutageneze. Na modelovém organismu Bacillus subtilis byla pomocí této metody m ena rychlost vzniku mutací v nestresovaném prost edí a v p ítomnosti n kolika vybraných stres . Bylo zjišt no, že limitace živinami ve stacionární fázi, hyperosmotický stres ani zvýšená kultiva ní teplota muta ní rychlost nezvyšují. Po sestrojení modelu pro m ení zm n exprese protein mismatch repair systému, nebyly p i porovnání míry exprese t... In a bacterium's environment, life conditions are subject to constant changes. One of the proposed mechanisms of adaptation to these changes is the increase in mutation rate. Bacterial mutability is generally kept very low by action of various mechanisms of control and repair, one of the most important ones being the Mismatch Repair, which is the master regulator of genetic stability of organisms. When its function is impaired, larger amounts of mutations occur in cells. In adverse conditions, these might be beneficial for cells' adaptation. The role of these repair mechanisms in adaptive processes in Bacillus subtilis has not yet been definitely resolved. The previous work in our lab focused on establishing an experimental system to measure the extent of mutagenesis in B. subtilis, and the influence of several stresses on mutation rate was assessed. No significant increase in mutability was found to be triggered by nutrient limitation in stationary growth phase, hyperosmotic stress or increased cultivation temperature. Furthermore, a system to monitor the expression of mismatch repair proteins was constructed, which has not revealed significant differences between stressed and nonstressed growth conditions.
