Anita Székelyová Vyhodnocení aktivity potenciálně antimikrobních látek pomocí mikrodiluční bujónové metody Rigorózní práce Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové Cílem této práce bylo stanovení antimikrobiální aktivity souboru 63 látek připravených na Katedře anorganické a organické chemie a Katedře farmaceutické chemie a kontroly léčiv Farmaceutické fakulty Univerzity Karlovy. Látky byly testovány mikrodiluční bujónovou metodou na 8 kmenech bakterií a současně na 8 kmenech patogenních hub. Testované kmeny zahrnovaly Gram-pozitivní i Gram- negativní bakterie včetně rezistentních původců nozokomiálních infekcí a také kvasinky i vláknité houby. Podle společných strukturálních znaků byly testované látky rozděleny do 7 skupin. Nejvyšší antibakteriální účinnost prokázaly deriváty 2-(5-karboxypentyl) isochinolinium- bromidu a 1-(5-karboxypentyl)pyridinium-bromidu, a to zejména na Gram-pozitivní bakterie. Měly také středně silný účinek na kvasinky. Nejvyšší antimykotickou aktivitu projevily deriváty 2-hydroxythiobenzamidu vůči Absidia corymbifera a látka WK-4 ze skupiny derivátů 2-[2-fenylvinyl]chinolinu, která dosáhla nízkých hodnot MIC téměř u všech testovaných kmenů, zejména u kvasinek. Anita Székelyová Evaluation of the activity of potentional antimicrobial substances through the use of microdilution broth method Rigorous thesis Charles university in Prague, Faculty of Pharmacy in Hradec Králové The aim of this thesis was to assess the antimicrobial activity of 63 substances prepared at the Department of Organic and Inorganic Chemistry and the Department of Pharmaceutical Chemistry and Drug Control, Pharmaceutical Faculty of the Charles University in Hradec Králové. The substances were tested through the microdilution broth method at eight strains of bacteria and pathogenic fungi at the same time. The tested strains consisted of Gram-positive and Gram-negative bacteria including highly resistant nosocomial pathogens, as well as yeasts and filamentous fungi. The tested substances were divided into seven groups according to their structural characteristics. The highest antibacterial activity was shown by the derivatives of 2-(5- carboxypentyl)isoquinolinium-bromide and 1-(5-carboxypenthyl)pyridinium-bromide, especially against Gram-positive bacteria. They also had intermediate acitivity against yeasts.
