[cze][eng] Tato diplomová práce je zaměřena na hledání bakteriálních původců a antagonistů ovlivňujících výskyt obecné strupovitosti, projevující se u některých zemědělsky využívaných plodin, jejímž hlavním původcem je Streptomyces scabiei a která způsobuje značné ekonomické ztráty. Z rhizosféry brambor byly izolovány bakteriální kmeny, především streptomycety, protože patří mezi nejvýznamnější producenty sekundárních metabolitů a původci tohoto onemocnění patří také do tohoto rodu. Metodou PCR byl u získaných kmenů amplifikován gen pro 16S rRNA a jeho sekvenací byla stanovena jejich příbuznost. Jednoduchým kultivačním testem byly poté nalezeny kmeny, které účinně potlačují S. scabiei. Dále byly metodou PCR hledány specifické geny pro biosyntézu fytotoxinu thaxtominu A, který je společným virulenčním faktorem všech doposud popsaných původců onemocnění a způsobuje projevy tohoto onemocnění na povrchu napadených plodin. Geny pro syntézu thaxtominu jsou součástí ostrovu patogenity. Standard fytotoxinu thaxtominu A byl použit pro optimalizaci jeho stanovení hmotnostní spektroskopií pro pozdější analýzu pokusů prováděných in vivo a in vitro. Výsledky fylogenetické analýzy izolovaných kmenů, u nichž byla prokázána přítomnost jednoho z genů nutných pro biosyntézu thaxtominu A, podpořily hypotézu horizontálního...This diploma thesis focused on screening for bacterial pathogens and antagonists suppressing common scab mainly caused by Streptomyces scabiei. Common scab affects some agricultural crops causing significant economical losses. Bacterial strains, mostly streptomycetes, were isolated from potato rhizosphere because they belong to most important producents of secondary metabolites and the causative agents of the disease are also members of this genus. The isolated bacteria were characterised by PCR amplification and sequencing of 16S rRNA gene to reveal their phylogenetic relationships. The ability of isolated strains to suppress growth of Streptomyces scabiei was tested by a simple co-cultivation experiment. The strains were tested by PCR for presence of specific genes for biosynthesis of thaxtomin A, a common virulence factor found in all described pathogens causing symptoms of this disease on the surface of affected tubers. Genes for synthesis of thaxtomin belong to pathogenicity island. Standard of phytotoxin thaxtomin A was used to optimize its analysis by mass spectroscopy for further in vivo and in vitro experiments.
