[cze][eng] Vitamíny jsou přírodní látky, které se nacházejí v rostlinách i živočiších. Až na několik výjimek si je lidské tělo nedokáže samo syntetizovat. Proto musí být dodávány potravou nebo potravinovými doplňky. Vitamíny jsou spolu s bílkovinami, tuky a sacharidy nezbytné pro růst, vitalitu, prevenci a léčbu mnoha zdravotních potíží a nemocí. Denní potřeba vitamínů je velmi nízká (jen několik miligramů či mikrogramů). Tudíž metody pro separaci či stanovení vitamínů např. z potravinových doplňků musí být vysoce citlivé. Mezi takové patří i vysokoúčinná kapalinová chromatografie s elektrochemickou detekcí. Diplomová práce se zabývá naleznutím optimálních podmínek pro separaci kyseliny listové, biotinu a vitamínu B12. Jedná se o vitamíny skupiny B, rozpustné ve vodě. Nejčastějšími zdroji je maso, vejce, pivovarské kvasnice nebo luštěniny. Při separaci byla použita chromatografická kolona Lichrospher 60, RP - select B (240 x 4,0 mm i.d., 5 μm) od firmy Merck. Analýza probíhala s využitím elektrochemického detektoru Coulochem III. Průtoková rychlost byla 1 ml/min a napětí na pracovních elektrodách bylo 600mV a 900 mV. Citlivost detektoru byla 1 μA. Byla zvolena mobilní fáze obsahující 24% methanolu. Anorganická fáze byla tvořena fosfátovým pufrem o koncentraci 0,05M. pH bylo upraveno na 3,55. PDF createdVYUŽITÍ HPLC V ANALÝZE LÉČIV II. Consequently, they must be supplied by the diet or by dietary supplements. Vitamins are essential for the normal growth, vitality, and for the prevention and cure of many health problems and diseases together with other essential nutrients such as proteins, fatty acids and carbohydrates. Vitamins are required only in tiny amounts by organism (micrograms or milligrams per day). Therefore, the methods for their separation and determination should be very sensitive. One possible method is the high performance liquid chromatography equipped with the electrochemical detection. This Diploma Thesis is focused on finding of optimal conditions for separation of folic acid, biotin and vitamin B12. These water-soluble substances belong to the B group vitamins. Their main sources are meat, eggs, brewer's yeast or pulses. The separation was carried out on chromatography column Lichrospher 60, RP - select B (240 x 4.0 mm i.d., 5 μm) obtained from Merck. The analysis was achieved by using the electrochemical detector Coulochem III with sensitivity of 1 μA.
