Cílem práce bylo zobrazit metodou mikroskopie atomárních sil (AFM) rozdíly ve struktuře buněčných stěn mladých a starých kvasinek S. cerevisiae a zjistit, jaký vliv na buněčnou stěnu má koncentrace glukózy v živném médiu. AFM je založena na měření sil mezi povrchem zkoumaného vzorku a hrotem, který povrch rastruje. Tato metoda umožňuje zkoumat buněčnou stěnu kvasinek v jejich přirozeném prostředí - ve vodě, přičemž lze zobrazit detaily o rozměrech řádově desítek nanometrů. Použity byly kvasinky kmenů AD 1-3,IL,US stejného stáří a kmenů JC 482A (označovány jako #013) a AD-JC 488A (označovány jako #015) staré jeden, dva a pět dní. Podařilo se nasnímat mnoho kvalitních obrázků srovnatelných s nejlepšími AFM snímky publikovanými v textech zabývajících se touto problematikou. Zejména se povedly detailně zobrazit oblasti jizev, které vznikají na povrchu kvasinek při jejich pučení a následném oddělení dceřinné buňky. Měření prokázalo výrazné změny buněčné stěny kvasinky v průběhu stárnutí (zvrásnění, zhrubnutí struktury). Vliv glukózy na strukturu buněčné stěny nebyl prokázán. Výsledky tohoto měření mohou být podkladem k dalšímu zkoumání buněčné stěny kvasinek metodou. The main goal of this MSc. is to use Atomic Force Microscopy (AFM) in studies of yeast cell wall structure aimed at (i) expected differences between young and aged S. cerevisiae cells, (ii) the effects of glucose concentration in culthuring media on the quality of yeast cell wall. AFM can image biological systems with molecular resolution in real time, and under natural condition. Numerous AFM images of the surface of S. cerevisiae yeast cells (strains AD 1-3,IL,US and JC 482A, AD-JC 488A) have been obtained, which surpassed that one of standard results of still-rare AFM studies of yeast cells. No clear effect of glucose concentration has been found. On the other hand, AFM studies have revealed ageing-related changes in the structure of yeast cells (crumpling, rough surface). Highresolution images of yeast bud scars, including their fine details were also obtained.
