Stále rostoucí rezistence vůči antibiotikům vede k potřebě nalezení nových účinných látek s antimikrobiálními vlastnostmi. Tato práce se zaměřuje na výskyt, chemický a fyzikální popis, mechanismus účinku a na biologickou aktivitu takovýchto látek, nalézajících se v jedu pavouků. Druhá část práce je zaměřena na izolaci a identifikaci látek s těmito vlastnostmi z jedu divokých včel a jednoho pavouka. Byl izolován a identifikován peptid s antimikrobiálními vlastnostmi a nízkou hemolytickou aktivitou z včely Trachusa byssina. Tento peptid má molekulovou hmotnost 1749,9 ? 0,1. Obsahuje 16 aminokyselin a je amidován na svém C-konci. Jeho primární struktura GILSVLKNLLKKHMAS-NH2 byla stanovena za pomoci Edmanova odbourávání a ESI-QTOF hmotové spektrometrie. Still increasing resistance to antibiotics leads to the need to find new active compounds with antimicrobial properties. This work is focused on the occurrence, chemical and physical description, mechanism of action and biological activity of such substances, found in spider venom. The second part is focused on isolation and identification of compounds with these properties from the venom of wild bees and a one spider. A novel peptide was isolated and identified from venom of bee Trachusa byssina. This novel peptide possess antimicrobial properties and low hemolytic activity. Molecular weight was estimated to 1749,9 ? 0,1contains 16 amino acids and is amidated on its C-terminus. Its primary structure GILSVLKNLLKKHMAS-NH2 was determined by using Edman degradation and ESI-QTOF mass spektrometry.
