Ateroskleróza patří mezi kardiovaskulární onemocnění, které tvoří v posledních letech nejčastější příčinu morbidity a mortality ve všech ekonomicky vyspělých zemích Evropy a USA. Cílem této diplomové práce bylo zjistit potencionální hypolipidemické a protizánětlivé účinky MDOC jako nové potenciální substance patří do skupiny dietární vlákniny. Ve studii byl použit geneticky modifikovaný kmen ApoE deficientní myši (n=8), které byly krmeny po odstavení aterogenní dietou s obsahem 1,25% cholesterolu po dobu 4 týdnů (kontrolní skupina). V MDOC skupině byly myši krmeny stejnou aterogenní dietou, ke které bylo přidáváno 50 mg/kg MDOC denně. Zvířatům byly odebrány ze srdce vzorky krve pro biochemické vyšetření. Dále byly odebrány segmenty tkáně tvořené aortou spolu s horní polovinou srdce. Dále byla provedena biochemická analýza krve a imunohistochemická a stereologická analýza endoteliální exprese VCAM-1 v aortálním sinu a oblouku. Výsledky biochemické analýzy ukázaly, že podávání MDOC neovlivnilo hladiny celkového cholesterolu, VLDL cholesterolu ani triacylglycerolů. Naproti tomu jsme zaznamenali statisticky významné snížení hladin LDL cholesterolu a rovněž statisticky významné zvýšení hladiny HDL cholesterolu. Imunohistochemická detekce exprese VCAM-1 byla pozorována převážně v endotelu aorty u obou skupin... Atherosclerosis belongs to the cardiovascular diseases, which have been the most frequent cause of the morbidity and mortality in all the economically advanced countries of Europe and the USA in the last few years. The purpose of this diploma dissertation was to find some potential hypolipidemic and antiinflammatory effects of MDOC. The new potential substance belongs to the group of dietary fibre. Genetically modified tribe of the ApoE-deficient mice (n=8) was used in the study. After the delactacion, it was fed on an aterogenic diet with 1,25% cholesterol content for 4 weeks. In MDOC group, the mice were fed on the same aterogenic diet, but just 50 mg/kg were daily added to this diet. Blood sample for the biochemical investigation were taken from animals. Then, partitions of the tissue formed by the aorta together with the upper part of the heart were taken, too. Then, a biochemical analysis of the blood was made and also in aortic sinus and arch an imunohistochemical and stereological analysis of the endothelial expression of VCAM-1 had to be done. The results of the biochemical analysis showed that serving of MDOC did not affect the total cholesterol level, VLDL cholesterol, or triglycerides.
