Autor: Kateřina Šumberová Název: Význam transformujícího růstového faktoru beta 1 a endoglinu v aterogenezi Diplomová práce Univerzita Karlova v Praze Farmaceutická fakulta v Hradci Králové Studijní obor: Farmacie Cíl: Cílem této diplomové práce je rešerše dostupných odborných poznatků o ateroskleróze, TGF - β1 (transformačním růstovém faktoru beta 1) a endoglinu a jejich roli v aterogenezi. Hlavní poznatky: TGF - β1 podporuje stabilitu aterosklerotického plátu, působí protizánětlivě a zabraňuje tak rozvoji a progresi aterosklerózy. TGF - β1 zároveň však omezuje regenerační schopnost endotelu, snižuje produkci vazodilatačních látek a naopak podporuje tvorbu vazokonstriktorů v cévní stěně. TGF - β1 podporuje vznik a rozvoj hypertenze, což přispívá k rychlejší aterogenezi. Endoglin je transmembránový protein, tvoří součást receptoru pro TGF - β a reguluje jeho signalizaci. Podle aktuálních poznatků se uvažuje o antiaterogenním působení endoglinu na cévní endotel. Závěry: Tyto výsledky naznačují, že role TGF - β1 a endoglinu v aterogenezi zůstává stále nejednoznačná. Pro určení přesných mechanismů a významů TGF - β1 a endoglinu v rozvoji a progresi aterosklerózy je nutný další výzkum. KLÍČOVÁ SLOVA: Ateroskleróza, TGF - β1, endoglin Author: Kateřina Šumberová Title of diploma thesis: Význam transformujícího růstového faktoru beta 1 a endoglinu v aterogenezi Diplomová práce Charles University in Prague Faculty of Pharmacy in Hradec Králové Field: Pharmacy Abstract Background: Objective of this diploma thesis is background research using available knowledge about atherosclerosis, TGF - β1 (Transforming growth factor - β1) and endoglin and about their roles in atherogenesis. Main findings: TGF - β1 promotes atherosclerotic plaque stability, acts as antiinflammatory agent and prevents from development and progression of atherosclerosis. However, at the same time TGF - β1 restricts endothelial regeneration, lowers production of vasodilators whereas TGF - β1 promotes production of vasoconstrictors in blood vessel wall. TGF - β1 supports initiation and progression of hypertension contributing to more progressive atherogenesis. Endoglin is a transmembrane protein, a component of the TGF - β receptor komplex and modulates its signalisation. According to recent studies is considered antiatherogenic action of endoglin to blood-vessel endothelium. Conclusions: These results demonstrate that the role of TGF - β1 and endoglin in atherogenesis remains still unclear.
