Abstrakt: [cze][eng] V teoretické časti práce byl zpracován přehled problematiky transdermální aplikace léčiv a s ní související problematiky propustnosti kožní bariery. Následně byly stručně shrnuty dosavadní poznatky o akcelerantech transdermální permeace, zejména pak transkarbamech a jejich vývoji na Farmaceutické fakultě Univerzity Karlovy. Na tuto část navazují výsledky permeačních pokusů prováděných na prasečí kůži in vitro. V nich byla hodnocena akcelerační aktivita různých koncentrací Transkarbamu 12 (T12) na permeaci kofeinu z oleokrémů s obsahem 60% vodné fáze. Pro krémy s 0%; 0,3%; 0,5%; 0,7% a 1% T12 byly zjištěny následující hodnoty průměrných fluxů kofeinu Jr: 7,88 ± 1,52 g.cm-2 .h-1 ; 9,34 ± 1,62 g.cm-2 .h-1 ; 10,89 ± 2,03 g.cm-2 .h-1 ; 11,29 ± 3,03 g.cm-2 .h-1 a 11,09 ± 2,21 g.cm-2 .h-1 . Present knowledge in transdermal permeation enhancers was then summarized shortly, with emphasis on Transkarbams and their development on the Faculty of Pharmacy of the Charles University. The results of the in vitro permeation experiments through full thickness pig ear skin are presented in the following experimental part. The acceleration effect of various concentrations of Transkarbam 12 (T12) on caffeine permeation from oleocreams containing 60% aqueous phase is evaluated. The following values of mean fluxes of caffeine Jr were found 7,88 ± 1,52 g.cm-2 .h-1 ; 9,34 ± 1,62 g.cm-2 .h-1 ; 10,89 ± 2,03 g.cm-2 .h-1 ; 11,29 ± 3,03 g.cm-2 .h-1 and 11,09 ± 2,21 g.cm-2 .h-1 for creams containing from creams containing 0%; 0,3%; 0,5%; 0,7% and 1% T12, respectively.
