Táto práca sa zaoberá využitím systému FLIM v konfokálnej mikroskopií. Popisuje základné pojmy, ktoré úzko súvisia s danou témou, ako je princíp fluorescencie, fluorescenčné značenie a podstata fluorescenčnej a konfokálnej mikroskopie. Praktická časť sa venuje práci s konfokálnym mikroskopom, na ktorom je zaobstaraná sada intenzitných snímkov rastlinnej a živočíšnej bunky. Konečným produktom praktickej časti bakalárskej práce sú snímky, na ktorých je v pseudofarbách vyobrazená doba života fluorescencie. V práci je tiež popísaný algoritmus k vytvoreniu pseudofarebných snímkov. K algoritmu je vytvorené uživateľské prostredie. This bachelor thesis deals with the FLIM system utilization in confocal microscopy. It describes basic concepts which are closely related to the topic, such as principles of fluorescence, fluorescent dyes and principles of both fluorescence and confocal microscopy. Practical part involves work with the confocal microscope and set of intensity images acquisition of both the animal and plant cells. Final product of pratical part is fluorescence lifetime image. The thesis also describes an algorithm for generating a pseudocolor frames and created user interface.
