Extraction of bioactive substances from black currant pomace

This bachelor thesis deals with extraction of bioactive substances from black currant pomace (Ribes nigrum). The theoretical part contains the botanical characteristics of the plant species Ribes nigrum. It is also devoted to the biologically active substances contained in black currant and possibilities of using black currant in the food industry. The fourth chapter deals with description of phenolic compounds, their distribution and methods of determination of phenolic substances. The fifth chapter describes selected extraction procedures. In the experimental part, there were extracted dry black currant pomace in order to optimize the extraction process and obtain the highest yields of dyes. In addition, were determined the selected chemical and physical characteristics of blackcurrant juice and extracts from blackcurrant pomace. A better solvent was a mixture of water and ethanol in a volume ratio of 1:1. After 14 hours maceration of dry pomace, were obtained 769,9 mg anthocyanins per 100 g of dry material.
Determination of basic chemical characteristics of blueberry pulp

This bachelor thesis deals with the investigation of selected chemical characteristics in the semifinished products of blueberries (Vaccinium myrtillus L.) and blueberries high (Vaccinium corymbosum L.). The theoretical part is described the botanical characteristics of both blueberries. The use of fruits is described in the next part. A description of the determination of the selected characteristic, refractometric of sugar dry mater, is described at the end of the theoretical part. In the experimental part, chemical characteristics were determined as total and soluble solids, pH, amount of titratable acids, reducing carbohydrates, phenolics and total anthocyanins in juice, pulp and extracts of blueberries. The purpose of the extracts was to find out how many total anthocyanins and phenolics are found in the skin of the fruit and which extract gives the highest yields. The results of the characteristics were compared across the wholefruit and semifinished products of the both blueberries. A mixture of water and ethanol in a ratio 50:50 was the most suitable solvent. The maximum extraction dependence was at twelfth and thirteenth hours of maceration. The maximum extraction dependence was determined by the above described chemical characteristics. Content of anthocyanin dyes expressed as delphinidin-3-glucoside was determined in extract of blueberry in 59,7 mg DGE100 g-1 and blueberries high in 19,6 mg DGE100 g-1. Total phenolics content expressed as gallic acid was determined in extract of blueberry in 890,9 mg GAE100 g-1 and blueberries high in 249,9 mg GAE100 g-1.
Possibility of further use of elderberry marc

This Diploma thesis talks about the production of concentrates and lyophilisate from the extract of elderberry moldlings (elderberry, Sambuvus nigra L.) and by determination by chosen chemical characteristics of these products. The theoretical part includes the botanical characteristics of the plant species Sambucus nigra L. Next is the part that is dedicated to biologicaly active substances found in elderberry and it‘s possible usage in the food industry. Next is the part where the extraction, purification and indentification of anthocyanic dyes are described. In the next part is the analyrical method HPLC described, including the description of instrumentation. The last part talks about the description of the used methods: methods for the concentration of the extract, UV-VIS spectrophotometry, determination of phenolic compounds, determination of dry matter, determination of active acidity and the overall titrating acidity. The experimental part contained the optimalization of the process of producing the extract from elderberry moldings. The extract obtained was then concentrated for preservation and characterization purposes based on selected chemical properties. The total content of anthocyanin dyes was chosen as a quality parameter. The most suitable process for the preparation of the extract was chosen to be the ratio of moldings and solvent 1:2, 4 (m/V). The most suitable solvent was chosen to be a mixture of water and ethanol in a ratio of 1:1 (V/V). According to the needs of the submitter, an extract with the use of water as the extraction agent is also provided. The prepared extract was then concentrated on a vacuum evaporator or lyophilized for storage purposes. The dye degradation while using the evaporator was the lowest in a 40 ° C water bath. The final products were characterized by the determination of soluble solids, pH, titration acidity, total phenolic compounds and total anthocyanin dyes. The anthocyanin pigments contained identified by liquid chromatography are: cyanidine-3-sambubioside, cyanidine-3-glucoside, cyanidine-3-sambubioside-5-glucoside, and cyanidine-3,5-diglucoside The total content of anthocyanins in the concentrate was determined to be equivalent to cyanidine-3-glucoside at 2.8 gl-1 and in the lyophilisate 2.5 gl-1.
Study of extraction of the biologically active substances from elderberry marc

This bachelor thesis deals with the determining of anthocyanin pigments in the extracts of elderberry marc (Sambucus nigra L.). The theoretical part is divided into four main chapters. In the first chapter there is botanical characteristics of elderberry, its origin and occurrence, chemical composition and the possibilities of usage in a food industry. The second part is devoted to characterization of phenolic compounds in berry fruits. Bigger attention is given to anthocyanin dyes, its occurrence, properties, health and antioxidant effects and the possibilities of usage in food industry. The third chapter is devoted to methods of determination of discussed dyes and phenolic compounds. The fourth chapter deals with the possibilities of disruption of plant cell walls. In the experimental part a series of elderberry marc's extraction was made. The purpose was to optimize the extraction course and to get the highest yield of dyes in an economically and operationally acceptable way. Time dependence of extractions was studied and a kind of extraction solvent and a ratio of its components were optimized. Distilled water or its mixture with ethanol was used as a solvent. During the water extraction the temperatures 25 °C and 50 °C were maintained and with the higher temperature the higher yields were obtained. Apart from this another supporting components as cellulolytic enzyme, ultrasound or shaker were used. The most effective method of extraction proved to be the extraction by the mixture water-ethanol (50:50) in the volume 80 mL. The total anthocyanins yield of this extraction was, expressed as cyanidin-3-glucoside equivalent, 914,4 ± 3,8 mg100 g-1 dry weight after 10 hours and the total phenolics yield was, expressed as gallic acid equivalent, 1957,0 ± 9,9 mg100 g-1 dry weight at the same time. The usage of ultrasound increased the efficiency of the extraction by water at 50 °C by 12 %. On the contrary, the usage of cellulolytic enzyme didn't increase the anthocyanin yield, because the enzyme was inactive.
Extraction of bioactive substances from black chokeberry pomace

This bachelor thesis deals with the quantification of selected bioactive substances in chokeberry juices, in the extracts of chokeberry and in chokeberry tea. In the theoretical part, there is a botanical description of chokeberry, (Aronia melanocarpa). Furthermore, the chemical properties of aronia and their use in the food, cosmetics industry and in the medicine. At the end of the theoretical part there are presented manual methods for determination of simple saccharides and briefly described selected extraction methods. The experimental part contains the determination of these chemical characteristics of analyzed aronia products: pH, titratable acidity, refractometric determination of dry matter, reducing sugars according to Bertrand's method and gravimetric determination, total phenolics and total anthocyanins. The highest phenolic content 319,54 mg·dm-3 were determined in a sample of aronia tea for 10 min digestion. The highest concentration of anthocyanin dyes was also determined in this tea – 889,49 mg·dm-3.
Extraction of anthocyanin pigments from elderberries pomace

This Diploma thesis deals with the production of the concentrate from the Elderberry marc extract (Sambucus nigra L.). The basic chemical characteristics in this concentrate were determined. The theoretical part of the thesis deal with the botanic characteristic of the Elderberry and its berries. The characteristic of the phenolic compounds, mainly anhocyanins was summarized. The use of the anthocyanin pigments in the food industry, the possibilities of their extraction and the concentrate production was discussed. Thereafter the methods of the anthocyanin content and the phenolic content determination were described. In the experimental part of study, the production of the anthocyanin rich extract from Elderberry marc was optimized. The extraction of the marc by the 1:1 ethanol/water (V/V) solvent with the ratio marc/solvent 1:2 (w/V) showed the most convenient. The production of the concentrate followed. During the concentration on the rotary evaporator the pigment degradation was investigated. The degradation decreased with the decrease of the temperature. The final concentrate was prepared from the extract on the rotary evaporator by 45 °C for 15 minutes. This product had the 7times lower volume compared to the initial extract. In the final concentrate come of the chemical characteristic were determined. By liquid chromatography (HPLC) the major pigments cyanidin-3-sambubiosid and cyanidin-3-glucosid were determined. Their total content was determined to 34,9 gl-1. The total anthocyanin content was determined by pH differential method to 7,1 gl-1 (expressed as cyanidin-3-glucoside equivalent). The total phenolic content was determined to 27,1 gl-1 (expressed as gallic acid equivalent) by Folin-Ciocalteu method. By electron paramagnetic resonance the increase in antioxidant activity with the increase in concentration was found. Up to 42 aromatic compounds was determined in Elderberry marc, its extract and the final concentrate.
