Methylation of viral RNA

Viruses are the major force that shapes the evolution of both pro- and eukaryotic organisms. They have a simple inner organization and contain only a few, usually well-described RNAs. In the case of +(ss)RNA viruses, their genomic RNA serves also as mRNA. This makes them a perfect model system for searching for new mRNA modifications as well as for understanding the role of already known modifications. In this work, Human Immunodeficiency Virus type 1 (HIV-1) from the Retroviridae family was used as a model system. In the following study, four representatives from the Picornaviridae family were tested for RNA methylation profile. To get the information, a combination of two techniques was developed, liquid chromatography- mass spectrometry (LC-MS) and sequencing techniques. Results of LC-MS reveal a surprisingly high amount of 1-methyladenosine (m1 A) in RNA isolated from HIV-1. Nevertheless, the m1 A mapping sequencing technique confirm m1 A position only in co-packed tRNA. This led to the recalculation of HIV-1 virion RNA composition. In the case of Picornaviridae, LC-MS revealed m1 A and 5-methylcytidine (m5 C) in two insect viruses (Sacbrood virus, SBV and Deformed wing virus, DWV). Bernard Malamud's Selected Fiction in the Context of Black-Jewish Literary Relations

Although Bernard Malamud's fiction has been frequently regarded as allegorical and symbolic, Malamud did not avoid the period's social issues in his works, such as the racial question and the changing nature of relationship between American Jews and African Americans. The present thesis aims to discuss Malamud's selected fiction dealing with Black- Jewish relations, namely short stories "Angel Levine," (1955) "Black Is My Favorite Color" (1963) and the novel The Tenants, (1971) and to place them into the context of Black-Jewish relations in the United States and of Black-Jewish literary dialogues and the tensions they express. It thus seeks to evaluate Malamud's role in the discourse of Black-Jewish relations in America. Calling upon a theoretical framework, outlined in chapter 2, based on philosophical and sociological findings of Judith Butler, John Searle, and Michael Omi with Howard Winant, the study examines the role of language and literature in constructing the Self and the Other (understood both as individual and collective identities, including categories of race and ethnicity), suggesting thus that literary texts, such as Malamud's selected fiction, are a part of discursive dialogue through and against which American Jews and Blacks construct their identities. New redox labels for DNA

The aim of my thesis was the synthesis of the modified 2'-deoxyribonucleoside triphosphates (dNTPs) bearing electrochemically oxidizable labels and their incorporation into DNA for the application in bioanalysis. In the first part of my thesis, I developed the synthesis of modified dNTPs bearing 2,3- dihydrobenzofuran (DHB) or 2-methoxyphenol (MOP) labels at 5-position of 2'- deoxycytidine 5'-O-triphosphate and at the 7-position of 7-deaza-2'-deoxyadenosine 5'-O- triphosphate by Suzuki-Miyaura cross-coupling reactions. Then modified dNTPs were used as substrates for DNA polymerases in enzymatic synthesis of modified DNA by PCR and primer extension. Electrochemical properties of the DHB and MOP-labeled nucleosides, dNTPs and DNA were studied by using of a square-wave voltammetry (SWV) at the pyrolytic graphite electrode (PGE) giving signals of MOP oxidation around 0.5 V and DHB oxidation around 0.85 V. The use of DHB group in combination with other electrochemical active labels was limited by close position of its oxidation peak to the signals of oxidation of natural nucleobases, whereas MOP moiety was successfully used for redox coding of nucleobases in combination with aminophenyl or benzofurazane label giving two independently readable redox signals in each case.
