Proteins demaged by oxidative stress, their role in physiology and ageing of yest cells
Mikešová, Jana ; Palková, Zdena (advisor) ; Borčin, Kateřina (referee)
Reactive oxygen species (ROS) are regularly produced in cells as a by-product of aerobic metabolism. Hence, organisms developed various defence mechanisms, which are able to avoid molecular damages caused by ROS under physiological conditions. In stress conditions, however, such defence mechanisms are not sufficient to avoid molecular damages. Accumulation of oxidized proteins is supposed to be a reason for ageing and many diseases including Friedreich's ataxia, Amylotrophic lateral sclerosis, Alzheimer's disease and many others. During oxidative stress, reactive oxygen species are reflected in oxidation of cystein residues in transcription factors, regulation proteins and active canters centers of enzymes. Oxidative modifications however could lead also to changes in transcription factor activity and activation of specific pathways, including changes in gene expression, cell cycle and proteolysis. This work shows defence mechanisms, ROS and proteins altered by reactive oxygen species that may function as important signalling molecules, which are essential for many cellular processes.
The influence of awareness about sexually transmitted diseases, sexual behavior of high school students
Mikešová, Jana ; Vachková, Eva (advisor) ; Hodačová, Lenka (referee)
Author: Jana Žalská Institution: Department of Social Medicine LF UK in Hradec Kralove Department of Nursing Title of the work: The influence of awareness about sexually transmitted diseases, sexual behavior of high school students Academic advisor: Mgr. Eva Vachková, Ph.D. Number of pages: 83 Number of attachments: 4 Defended in: 2018 Key words: adolescence, sexually transmitted diseases, sex, sexual education, health literacy, gender identity The bachelor thesis is on the topic "Influence of awareness of sexually transmitted diseases on the sexual behaviour of high school youth". The theoretical part deals with sexual diseases and sexually transmitted diseases. It describes the period of adolescence, physical transformation and gender differences in adolescence. It also deals with health literacy in youth, health education, and sexual education. The empirical part looks into the knowledge of secondary school youth about sexual diseases and their prevention and the way of protection against sexually transmitted diseases, next this part looks into the sources from which they receive information. It also investigates whether their knowledge of this issue has an effect on sexual behaviour. Quantitative research was applied by a non-standardized questionnaire. The resulting data is processed into...
Sch9p and Rim15p signaling pathways in yeast colony development
Mikešová, Jana ; Palková, Zdena (advisor) ; Heidingsfeld, Olga (referee)
On solid surfaces, laboratory strain Saccharomyces cerevisiae BY4742 forms horizontally and vertically stratified colonies. Central part of the colony differentiates into U and L cells occupying upper and lower colony regions, respectively. There are many morphological and physiological differences between these two cell types. However, molecular mechanisms of the vertical stratification are still largely unknown. In this study, I analyzed effects of the deletion and overexpression of Sch9p a Rim15p kinases with crucial role in nutrient sensing pathways, of transcription activators Msn2p and Msn4p involved i stress response and of post-diauxic shift transcription factor Gis1p in yeast colony differentiation and U and L cell survival. Microscopic analyses showed changes in morphology of cells in colonies of these strains and decreased ability of vertical stratification in colonies of the strain rim15Δ and partially in colonies of the strain PTEF-SCH9. Moreover, changed level of these proteins significantly affected long-term survival of U and L cells. Obtained data suggest an involvement of nutrient sensing pathways in colony formation and development.

Activating methods in business education
Mikešová, Jana ; Krpálek, Pavel (advisor) ; Krpálková Krelová, Katarína (referee)
The diploma thesis Activating methods in business education is focused at training Prince project management for the company Symphera. The aim is to create methods for activation of Prince2 training and incorporate them into new methodologies of training. In the theoretical part we will introduce various activating methods. We will explain what they mean and what training methods are used within them. In the part Creation of activation methods we will describe what was the process of working on the inclusion of activating methods in the training course of Prince2 project methodology. We will review the selection and subsequent formation of individual methods. The aim of comparing the success of the course is to determine whether students who have completed a course of activating methods achieve better test results than those who completed the course the classic method.
System proposal of further education in business sector
Mikešová, Jana ; Krpálková Krelová, Katarína (advisor) ; Kolářová, Dana (referee)
The aim of bachelor's thesis "System proposal of further education in business sector" is the education of employees in KBC Global NV Czech Branch. The objective of this proposal is to design system which would be meant to advance the quality of employees' education in given company. In theoretical part we will look closer at the meaning of adults' education. In analysis of the system, there is described chat system of education KBC Global NV Czech Branch uses. The main objective of quantitative research is to find out what is the level of employees' satisfaction with educational system in KBC Global NV Czech Branch, what forms of educational system in given company the employees are fully satisfied with or not and not least chat changes they would like to make, if any. The optimization proposal is focused on more efficient educational system which would advance the quality of education in KBC Global NV Branch and therefore to gain higher value of human capital.
