Emperor Rudolf II's Patronage of Alchemy. Michael Maier (1569-1622) and Anselmus Boëthius de Boodt (1550-1632) The work is focused on the emperor Rudolf II's (1552-1612) patronage of alchemy and two distinguished physicians interested in alchemy that lived for some time at the emperor's court. The text reflects these topics in its structure; therefore, it is divided into three parts. The first part deals with the emperor himself and his support of alchemy at the court. Furthermore, the reasons of the emperor's interest in this field of research are analyzed, as well as historical and philosophical context of the research. Examined were also collaborators and closest courtiers to the emperor that took and interest in alchemy and the possibilities of practical realization of laboratory works at Prague castle, the question of their nature and expected results. Last but not least is discussed, to what extent was the emperor personally involved in the experiments. The second chapter is about Michael Maier (1569-1622), a physician and alchemist, who came to Prague in the middle of 1608 with an intention to ask the emperor to support his research. Maier gained a doctorate in medicine in Basel and his interest in alchemy was purely medical. He wanted to offer the emperor a 'universal medicament' he had...
Catalytic and adsorption properties of papain and its derivatives

The aminoacid sequence of papain (EC 3.4.22.2) consists of 212 aminoacids. It has only one free sulfhydryl group, which is located in the active site of the protein. Some organometallic complexes could be bonded only to this free -SH group due to their structure. The artificial metalloproteins synthesised by this way may have different electrochemical properties. In this work, we have studied the electrochemical properties of papain and its derivatives. We compared the ability of papain and its three artificial derivatives to catalyse the hydrogen evolution by the chronopotenciometry. The work was completed by the study of the electrochemical properties of the organometallic complexes of ruthenium, which were used for the artificial metalloprotein preparation. The electrochemical properties of the compounds were never studied before. The process of the hydrogen evolution catalysed by the proteins is held in the adsorbed state of the catalyst. Due to this fact we have also studied the adsorption properties of papain on the substrates with different level of hydrofobicity. (In Czech)
Role of protein-protein interactions in regulation of signalling proteins and enzymes

EN Protein-protein interactions have an exceptional position among other mechanisms in the regulation of signal transduction. Their systematic investigation is very important and logical step in the process of understanding to the transduction and its mechanisms at a molecular level. During my Ph.D. I was particularly interested in three important processes. ASK1 kinase is well-known initiator of the apoptosis. Under physiological conditions it is maintained in an inactive state by its two interaction partners the 14-3-3 protein and TRX1. These two proteins dissociate in the presence of reactive oxygen species by unclear mechanism and the kinase is therefore activated. The next process is an interaction between the 14-3-3 protein and phosducin and investigation of their role in the G protein signalling especially important in the biochemistry of vision. The third process is an activation of protein Nth1 through the interaction with Bmh1, yeast analog of the 14-3-3 protein, and calcium cations. I employed various biophysical method, particularly analytical ultracentrifugation, in order to explain molecular mechanisms of described processes. Integrated Science Education and History of Science in Chemistry Education

The present dissertation Integrated Science Education and History of Science in Chemistry Education is aimed at the problem of integrating scientific subjects by means of natural history. The thesis contains research concerned with integrating science education in its various forms and its analysis from the point of valid curricular documents. Results of a questionnaire on intersubject integration distributed among grammar school chemistry teachers are presented in the thesis. Further on, an approach to the integration by means of natural history is presented. Didactic materials were created on this topic: worksheets and didactic tests in Adobe Flash. Worksheets were attested in grammar school teaching and results of this attestation are included. The thesis also includes results of two projects aimed at the topic of natural history. These projects are: History of Natural Sciences in High School Teacher Education (Fund for the development of higher education in the Czech Republic, 2011) and a course called Alchymistic Prague (Developmental project of the Ministry of Education, 2011: Chemistry year - Chemistry for all).
