Timber framework of family house in Protivín

The subject of this thesis is the analysis of load bearing timber structure of a family house. The house is designed as two-floor building. The plan dimension of the building is 11x8m and the height is 7m. The load bearing structure of the roof is designed by timber counter truss. The load bearing structure is designed from timber columns with dimensions 60 x 120mm, which form a spatial system in plan grid 625mm. The house is situated in South Bohemia in the city Protivín.
Development of the construction of residential properties in České Budějovice

The aim of the theoretical part of my Master’s thesis is to summarize the historical evolution of building construction in České Budějovice, to process the summary of residential construction in each area of the city, and to describe current construction. There is described in my thesis how was the city of České Budějovice developing, varying and ramifying over time. In the practical part I did the analysis of the future construction using the survey among residents of České Budějovice and so future builders. Analysis determines the requirements for the type and location of the future construction, builders’ own building requirements. This thesis summarizes and describes the history and current construction. Also thanks to the practical part gives awareness about the direction of residential constructions in České Budějovice in the coming years.
Leadership in the management of organizations providing social services.

The thesis on Leadership in the Management of Social Services Organizations focuses on social service managers and explores how they strengthen and develop leadership skills. Leadership is important in social service management because it allows for effective team management focused on achieving common goals and improving the quality of services provided to users. While some managers may rely primarily on their abilities and the application of leadership skills may be more intuitive, others may consciously apply specific leadership skills. The theoretical part of the thesis clarifies the concepts of management and leadership and describes the difference between managers and leaders. These concepts are then related to the context of social services and the topic of leadership skills and how they can be applied in the management of social services is further developed. The empirical part of the thesis aims to answer the question "How do managers in the practice of social services strengthen and develop leadership skills?" Qualitative research is used and respondents are line managers in social services who will undergo a semi-structured interview. The life quality of patients with the permanent cardiac stimulator

Key words: life quality, permanent cardiac stimulator, patient This bachelor thesis is aimed at the problems of the life quality of the patients with the cardiac stimulator. The theoretical part of this work deals with the pieces of knowledge about the people with implanted permanent cardiac stimulator and their life quality. In the first chapter there is the description of anatomy and the physiology of cardiovascular system. The next one deals with the problems of cardiostimulation. The third chapter is focused on nursing care before and after the implantation. Thereinafter the education of the patiences is described. The important chapter in the theoretical part is about the life quality. The last point mentioned is the introduction with the notion home monitoring. The practical research was done to discover if there was a life change after the implantation in the patients'lives. The anonymous nonstandard questionnaire was used for this research. A few questions were used from the questionnaire LHFQ (Living with heart failure questionnaire) . The research was done from December 2007 to March 2008 in the cardiology consulting room and in internal sick-bed ward of the hospital in Havlickuv Brod.
Cysteine tRNA regulates protein synthesis in human cell lines

A significant number of known human genetic diseases is associated with nonsense mutations leading to the introduction of a premature termination codon into the coding sequence. A termination codon can be read through by its near-cognate tRNA (tRNA with two anticodon nucleotides base-pairing with a stop codon); potentially generating C-terminally extended protein variants. In yeast, UGA stop codon was described to be read through by tRNA-Trp and tRNA-Cys. Similar was observed for tRNA-Trp in human HEK293T cell line. The aim of this thesis was to investigate if human tRNA-Cys can act as a near-cognate tRNA in human HEK293T cell line. There are two isoacceptors which constitute the tRNA-Cys family, with ACA and GCA anticodon. There are 1 and 23 isodecoders to the ACA and GCA anticodons, respectively. Here, altogether as many as nine tRNA-Cys isodecoders (distinct in their sequence and with varying levels of expression) were tested for their ability to increase UGA readthrough in HEK293T using p2luci and pSGDluc dual-luciferase reporter vectors. In both p2luci and pSGDluc, we observed that at least one tRNA-Cys isodecoder, tRNA-Cys-GCA-4-1, is capable of significantly elevating the UGA readthrough levels when overexpressed in HEK293T. miRNA-mediated translational repression

Recent models of miRNA-mediated translational repression favor initiation as the phase of translation, where the repression is primarily evoked. miRNA-induced silencing complex utilizes miRNA as a guide molecule to find its target mRNA. To inhibit mRNA translation and induce its decay, many other proteins are then recruited, e.g. GW182 protein, PAN2-PAN3 a CCR4-NOT deadenylation complexes, decapping enzyme and eIF4E-binding proteins. miRNA-mediated translational repression is then established by either their concerted action or their consecutive and coordinated binding to target mRNA. To date, several mechanisms have been described: GW182- mediated PABP displacement; GW182-independent translational repression; miRNA-mediated eIF4A1 dissociation or eIF4A2 recruitment; translational repression mediated by deadenylases CAF1 and CCR4; and 4E-T, eIF4E2, GIGYF2 translational repressor proteins recruitment. Here, I provide up-to-the-minute summarization of these mechanisms with the aim to extract any universality from them.
Farmer's reasons to engage in organic agriculture

The master thesis deals with farmer's reasons to engage in organic agriculture. The theoretical part is devoted to the topic of organic agriculture, its development in the Czech Republic and abroad, organic farming legislation and subsidy policies in the Czech Republic. Described methodology is a part of master thesis. The empirical part provides the readers with the presentation of eight individual organic farms in the regions of Hana and Beskids. On the grounds of empirical survey carried out through in-depth semi-structured interviews, the thesis reveals inner motivations that stimulate farmer's commitment to work in an environment-friendly way. Then, detailed description and interpretation of the codes resulting from open coding of the interviews are presented.
Variants of knapsack cryptosystems

The topic of this thesis is a cryptosystem, precisely a public key encryption scheme, that is based on the knapsack problem. At first we formulate terms like \mathcal{NP} -complete problem, one-way function, hard-core predicate, public key encryption scheme and semantic security which we connect in this thesis. After that we present the knapsack problem. Then we prove that the knapsack problem with appropriate parameters has a property that leads to semantic security of the encryption scheme which we present afterwards.
