Epidemiological studies suggested that there is a low incidence of breast cancer in countries with high soya intake. Soya contains the isoflavones daidzein (D) and genistein (G), which are responsible for the protective properties, although their exact effects have not yet been clarified. In this study we evaluated and compared the biochemical and biological effects of a soya extract biotransformed by Aspergillus Awamori (SBE) and of the pure major isoflavones, G and D in placental microsomes and in an estrogen-dependent breast cancer cell line stably transfected with the aromatase gene, the MCF-7aro cell line. The results showed that D did not induce marked alterations in any of the parameters studied. In placental microsomes G was not a potent aromatase inhibitor, inducing only a moderate reduction in the aromatase activity. In MCF-7aro cells it was observed a significant...
Acidic Hydrolysis of Waste Feather Biomass - Processes Scale-Up.
Hanika, Jiří ; Rousková, Milena ; Šolcová, Olga ; Šabata, Stanislav ; Jírů, M. ; Hůrková, K.
The paper present novel data on acidic hydrolysis of waste chicken feathersin presence of water solution containing carboxylic acid catalyst with the pKa value lower than 4, instirred batch reactors, in the temperature range from 114°Cto 150°C and at the pertinent vapour pressure of reaction mixture. The process was scaled-up from laboratory bench scale to pilot plant dimensions using batch high pressure autoclaves. Process conditions were applied according to patent application1. Knowledge of kinetic aspects of the featherhydrolysis and process parameters operation window are inevitable for asuccessful process scale up2,3.

Simultaneous Hydrolysis of Proteins and Fats fro Waste Chicken Feathers.
Rousková, Milena ; Šolcová, Olga ; Šabata, Stanislav ; Jírů, M. ; Hůrková, K. ; Hanika, Jiří
The kinetics of the simultaneous hydrolysis of macromolecular proteins (keratin) and lipids (fat) from waste chicken feathers was investigated in this study. The perspective option pressure hydrolysis at elevated temperature was designed as one of the primary tasks within the research program in frame of the Competence Centre BIORAF, supported by the Technology Agency of the CR. The process development of simultaneous hydrolysis of proteins and lipids contained in the waste is focused on this potential alternative raw material for the preparation of valuable amino acids1 and proteins of low molecular weight. In this study, the acidic hydrolysis kinetics of the waste biomass was performed using a batch stirred bench scale autoclave at elevated temperature and pressure of inert atmosphere from 0.9 to 2.3 MPa. Produced hydrolysate was filtrated and subsequently analyzed using HPLC/MS method at the UCT Prague.

Fatty Acids Profile Analysis in Fresh Suspension of Japonochytrium Sp.
Rousková, Milena ; Maléterová, Ywetta ; Hůrková, K. ; Kaštánek, P. ; Hanika, Jiří ; Šolcová, Olga
The study is focused on concentration of a lipid content and determination of the extractable part oriented on included fatty acids in separated phases of centrifuged wet biomass. The main attention was focused on the DHA concentration from the upper lipid layer, containing obviously concentrated proportion of fatty acids. Water fungi suspension was submitted to centrifugation with subsequent separation of individual liquid/solid fractions and extracted by two solvent systems. Dichloromethane/methanol 1/1 and 1-hexane/ethanol 2/3 was applied. Both solvent systems afforded comparable fatty acids profiles. Dominant acids C16:0 (in the range of 43-47%) and DHA C22:6n3 (43-47%) were detected. Biomass residue can be applied e. g. as an animal food supplement due to high protein content. Also level of pigments with antioxidant effect, particularly carotenoids, can be dietary significant.

Supercritical Fluid Extraction: A Green Method for the Isolation of Valuable Substances from the Various Biological Matrices.
Topiař, Martin ; Sajfrtová, Marie ; Šolcová, Olga ; Hradecký, J. ; Hůrková, K. ; Hajšlová, J.
Supercritical fluid extraction (SFE) using carbon dioxide is a sophisticated technology suitable for isolation of un-polar, high value biological active substances from the various biological matrices. In this study, it was used for the extraction of Magnolia x pruhoniciana flowers and Japonochytrium marinum fungus. Results obtained from the SFE were compared with those from conventional extraction methods. A total number of 37 compounds were identified in the magnolia isolate, from which α-pinene (18 % of peak area), cymene (14 % of peak area), β-pinene (13 % of peak area) and humulene (6 % of peak area) were its main components. Yield of valuable free fatty acids DHA and EPA obtained by the SFE of Japonochytrium marinum was more than 1.5 times higher in comparison with organic liquid extraction using mixture of methanol and dichloromethane.
