Title of rigorous thesis: Study of renal transport mechanisms of radiolabeled antibodies Utilization of monoclonal antibodies belongs to one of the important strategies in the treatment of oncological diseases. This treatment has a great advantage in precise focusing on the target structure with minimal damage to surrounding non-target tissue. The antibodies against the receptor for epidermal growth factor (EGFR) expressed in higher rate by variety of tumor types represent one of the important group. Nowadays oncology utilizes both unconjugated antibodies, and the ones conjugated with other effector components, e.g. cytostatic agents, radionuclides, toxins. One of the important limits for the antibodies conjugated with radionuclides appears to be their potential accumulation in kidneys with subsequent radiotoxic damage to the kidney tissue. Evaluation of oxime cholinesterase reactivators by neurobehavioral methods

Na albinotickych samcich potkanů kmene Wistar byla hodnocena schopnost oximovych reaktivatorů enzymu acetylcholinesterazy (AChE, EC 3.1.1.7) eliminovat projevy neurotoxicity navozene nervově paralytickymi latkami in vivo. Ve dvou pokusech byl zviřatům podan i.m. tabun, ve třetim soman, vždy v davce 80 % LD50 a po uplynuti 1 minuty byla podana i.m. terapie obsahujici anticholinergikum atropin (21 mg/kg) vždy v kombinaci s jednim z vybranych oximů. Do pokusů byly zařazeny nově syntetizovane latky K074 a K075, dale pak obidoxim, trimedoxim a HI-6 jako srovnavaci latka. Všechny testovane latky byly v průběhu všech pokusů aplikovany v ekvimolarnich davkach (50 mol/kg). Pro hodnoceni jednotlivych projevů tabunem navozene neurotoxicity byla použita metodika Funkčni pozorovaci baterie, ktera obsahuje hodnoceni senzorickych, motorickych a autonomnich nervovych funkci. Jednotlive ukazatele navozene neurotoxicity budou hodnoceny 24 hodin a 7 dni po intoxikaci a porovnavany s daty kontrolni skupiny, ktere byl i.m. podan namisto nervově paralytickych latek a antidot fyziologicky roztok. Cilem tohoto souboru pokusů je prokazat pozitivni terapeuticky efekt testovanych reaktivatorů AChE a hodnotit parametry učinnosti jednotlivych terapeutickych kombinaci. This is because determining the concentration of the drug of interest in biological matrices can help to reveal its mechanism of action, determine the values of basic pharmacokinetic parameters or study the fate of a potential drug during in vitro experiments. Considering the complexity of biological samples and low concentrations of studied compounds, the sample preparation prior to analysis is really important. Miniaturized techniques derived from standard extraction methods have significant presence in modern bioanalysis, mainly because they decrease both required sample volume and consumption of laboratory material to a minimum, and they enable automatization. One of the main areas of drug discovery is currently the treatment of cancer. Unfortunately, development of highly efficacious drugs and prolongation of patient survival have resulted in the risk of developing late complications associated with anticancer treatment...

Nature field trip from a teacher's point of view

Thesis is focused on all aspects of having pupils of a younger school age participating in a nature field trip, as percieved from a teacher's point of view. The theory section concentrates on the definition of basic terms on given issues, legislation and organisation of the nature field trip. The practical part is formed of a questionnaire section and a discussion with the aim to find out whether schools in the Czech Republic organise these trips and if so, what experiences teachers come across. If not, what reasons let them to that decision. The object of the thesis is to analyse the outcome of the inquiry and to summarise the benefits and disadvantages which emerge for teachers from such nature field trip. Both part sof the thesis highlight the challenges teachers are facing in thein profession not only during the preparation stages but also throughout the course of the nature field trip. Additionally the practical section features actual examples of previously completed trips, including the reflection of participating teachers.
